摘要
肿瘤细胞代谢与正常细胞不同,为了获得其快速增殖能力所需的物质和能量,肿瘤细胞能量代谢从有氧氧化转化为有氧酵解。3-磷酸甘油酸脱氢酶(PHGDH)作为一种参与丝氨酸从头合成的关键酶,在肿瘤细胞增殖、转移过程中发挥重要作用,沉默PHGDH基因可导致肿瘤细胞增殖减少,凋亡增加,耐药性减弱。PHGDH参与乳腺癌、肺癌、黑色素瘤等肿瘤的发生发展过程,其高表达与患者生存率下降、远端转移以及肿瘤耐药等相关。未来,PHGDH有望成为肿瘤治疗的新靶点。
The metabolism of tumor cells is different from normal cells.In order to adapt to its rapid proliferation ability, the energy metabolism of tumor cells converts from aerobic oxidation to aerobic fermentation.As a key enzyme involved in serine de novo synthesis, 3-phosphoglycerate dehydrogenase(PHGDH) plays an important role in the proliferation and metastasis of tumor cells.Silencing PHGDH genes lead to reduced tumor cell proliferation, increased apoptosis and weakened drug resistance.PHGDH is involved in the occurrence and development of various tumors such as breast cancer, lung cancer and melanoma.High expression of PHGDH is associated with clinical manifestations such as decreased survival, distal metastases, and tumor resistance.In the future, PHGDH is expected to become a new target for the clinical tumor treatment.
