摘要
目的:旨在揭示纺锤体与动粒相关蛋白3(SKA3)在肺腺癌中的表达、潜在的临床价值及其可能的调节机制。方法:利用癌症基因组图谱(TCGA)数据库RNA表达数据分析SKA3在肺腺癌中的表达,Wilcoxon秩和检验和逻辑回归分析肺腺癌患者的临床病理学特征与SKA3之间的相关性。Cox回归和Kaplan-Meier生存分析与预后相关的临床病理学特征,并对TCGA数据库的基因集进行富集分析(GSEA)探究SKA3调节肺腺癌进展的可能机制。采用qRT-PCR检测肺腺癌组织中SKA3mRNA表达,分析其表达水平与肺腺癌患者临床病理学特征之间的关系。结果:SKA3在肺腺癌组织中高表达。SKA3高表达与肺腺癌患者年龄、性别、临床分期、T分期、淋巴结转移和预后显著相关(P<0.05)。单因素分析和多因素分析显示SKA3表达水平是肺腺癌患者预后不良的独立危险因素(P<0.05)。SKA3高表达组显著富集到细胞周期、DNA复制和p53信号通路等。结论:SKA3在肺腺癌中高表达,SKA3高表达与肺腺癌发生发展密切相关,可能是肺腺癌预后不良的潜在分子标志物,SKA3可能通过调控细胞周期、DNA复制和p53信号通路促进肺腺癌进展。
Objective:To reveal the expression of Spindle and kinetochore associated complex subunit 3(SKA3)in lung adenocarcinoma,its potential clinical value and its possible regulatory mechanism.Methods:The expression of SKA3 in lung adenocarcinoma was analyzed by RNA expression data of the Cancer Genome Atlas(TCGA),and the correlation between clinicopathological features and SKA3 in patients with lung adenocarcinoma was analyzed by Wilcoxon rank sum test and logical regression.Cox regression and Kaplan-Meier survival analysis were used to analyze the clinicopathological features related to prognosis,and the gene set of TCGA database was analyzed by GSEA to explore the possible mechanism of SKA3 regulating the progression of lung adenocarcinoma.The expression of SKA3 mRNA in lung adenocarcinoma was detected by qRT-PCR,and the relationship between the expression level and clinicopathological features of lung adenocarcinoma was analyzed.Results:The expression of SKA3 was increased in lung adenocarcinoma.The increased expression of SKA3 is significantly correlated with age,sex,clinical stage,T stage,lymph node metastasis and prognosis of patients with lung adenocarcinoma(P<0.05).Univariate and multivariate analysis showed that the expression of SKA3 was an independent risk factor for poor prognosis in patients with lung adenocarcinoma(P<0.05).SKA3 high expression group was significantly enriched in cell cycle,DNA replication and p53 signal pathway.Conclusion:The high expression of SKA3 and SKA3 in lung adenocarcinoma is closely related to the occurrence and development of lung adenocarcinoma,and may be a potential molecular marker for poor prognosis of lung adenocarcinoma.SKA3 may promote the progression of lung adenocarcinoma by regulating cell cycle,DNA replication and p53 signaling pathway.
