摘要
目的:探讨重灸法对失眠模型大鼠下丘脑Orexin-A表达的影响。方法:将40只SD大鼠随机分为空白组、模型组、重灸组、针刺组和温和灸组,每组8只。除空白组外,其余组大鼠以0.3 g·kg^(-1)的剂量腹腔注射对氯苯丙氨酸溶液,每天1次,连续2 d,建立失眠大鼠模型。建模成功后,重灸组大鼠于背俞穴一次性灸至破溃或化脓,针刺组大鼠于背俞穴进行揿针治疗,温和灸组于背俞穴处进行常规温和灸治疗,空白组、模型组大鼠只固定不做其它处理,干预6天。分别采用免疫组化法和Western Blot法检测下丘脑组织中Orexin-A的表达。结果:与空白组比较,模型组大鼠下丘脑Orexin-A的表达显著升高(P<0.05);与模型组比较,重灸组、针刺组、温和灸组大鼠下丘脑Orexin-A的表达显著降低(P<0.05);与重灸组比较,温和灸组与针刺组大鼠下丘脑Orexin-A的表达显著升高(P<0.05)。结论:重灸法可明显抑制失眠模型大鼠下丘脑Orexin-A的表达。
Objective:To observe the effect of heavy moxibustion on expression of Orexin-A in the rat model of insomnia,so as to provide experimental evidence for treating insomnia in clinical treatment.Methods:Forty SD rats were equally randomized into blank group,model group,heavy moxibustion group,acupuncture group and mild moxibustion group(n=8 rat/group).Except for the blank group,rats in the other groups were intraperitoneally injected with p-chlorophenylalanine solution at a dose of 0.3 g/kg,once a day for 2 consecutive days,to establish a rat model of insomnia.After successful modeling,rats in the heavy moxibustion group were moxibustion at the Bei Shu acupoint at one time until they became ulcerated or suppurative;the rats in the acupuncture group were treated with acupuncture at the Backshu point,and the mild moxibustion group was treated with conventional mild moxibustion at the Bei Shu acpoint,The rats in the blank group and model group were only fixed without other treatments,and intervened for 6 days.The expression of Orexin-A in hypothalamic tissue was detected by immune-histochemical method and Western Blot method.Results:Compared with the blank group,the expression of Orexin-A in the hypothalamus of rats in the model group was significantly increased(P<0.05);Compared with the model group,the expression of Orexin-A in the hypothalamus of rats in the heavy moxibustion group,acupuncture group,and mild moxibustion group was significantly reduced(P<0.05).Compared with the heavy moxibustion group,the expression of Orexin-A in the hypothalamus of the mild moxibustion group and the acupuncture group was significantly increased(P<0.05).Conclusion:Heavy moxibustion can significantly inhibit the expression of Orexin-A in the hypothalamus of insomnia model rats.
