质控图在荧光定量PCR检测室内质量控制中的应用研究

Application of Quality Control Chart in Laboratory Quality Control of Fluorescent Quantitative PCR

针对荧光定量PCR仪的温度性能及扩增质量,本文采用PCR仪无线温场校准装置及大肠埃希氏菌EHEC DNA标准品作为质控品,对PCR仪的扩增温度及扩增质量进行测试,并绘制质控图。结果表明质控图能够连续、动态地反应同一台荧光定量PCR仪若干次试验的稳定性及扩增质量。通过绘制质控图对实验结果和扩增温度进行质控,能尽早发现扩增异常的实验批次或温度产生漂移的设备,保证于弱阳性样品的检出,避免漏检、误检。

In view of the temperature performance and amplification quality of fluorescence quantitative PCR instrument,the wireless temperature field calibration device of the PCR instrument and Escherichia coli EHEC DNA standard were used as quality control products to test the amplification temperature and quality of the PCR instrument,and the quality control chart was drawn.The results showed that the quality control chart could continuously and dynamically reflect the stability and amplification quality of several experiments with the same fluorescent quantitative PCR instrument.By drawing the quality control chart for the quality control of the experimental results and amplification temperature,the abnormal amplification of experimental batches or equipment with temperature drift can be found as soon as possible,so as to ensure the detection of weak positive samples and avoid missed detection and misdetection.
