Research progress on the role of fibroblast growth factor 21 in pathogenesis of alcoholic liver disease

成纤维细胞生长因子21(fibroblast growth factor 21,FGF21)是一种主要由肝脏合成分泌的内分泌激素,既往研究提示其在人体糖、脂肪与能量代谢中发挥重要调节作用。酒精性肝病是危害人类健康的主要肝脏疾病之一。近年来研究提示FGF21在酒精性肝病致病过程中发挥多种保护作用。FGF21不仅可以下调机体饮酒欲望,改善肝脏脂肪变、肝脏炎症及纤维化,还可在肝癌的发生中发挥一定保护作用。目前,FGF21用于糖尿病与代谢相关脂肪性肝病的临床试验正在进行中,FGF21作为酒精性肝病的治疗靶点值得深入研究。

Fibroblast growth factor 21(FGF21)is a hormone which is mainly synthesized in liver.Previous studies showed that FGF21 plays important roles in regulating carbohydrate,fat and energy homeostasis.Alcoholic liver disease(ALD)is one of the most common liver diseases which impair people's health.Recent studies show that FGF21 plays multiple protective roles in pathogenesis of ALD.FGF21 can reduce the alcohol preference,attenuate hepatic steatosis,inflammation,fibrosis,and decrease risk of liver cancer.Clinical trials of FGF21 for diabetes mellitus and metabolism associated fatty liver disease are going on.Further studies on FGF21 as the therapeutic target of ALD are needed.
