摘要
为探究ANKRD17基因g.88739493 T>G位点和CLEC16A基因g.9587011 G>A、g.9730880 G>A位点多态性及其与山羊繁殖性能之间的关联,利用Sequenom MassARRAY^(■)SNP分型技术对云上黑山羊、济宁青山羊和辽宁绒山羊3个群体ANKRD17和CLEC16A基因的3个位点进行了多态性检测,并与云上黑山羊繁殖性能(包括产羔数、初生窝重和断奶窝重)进行关联分析。结果表明,ANKRD17基因g.88739493 T>G在3个群体中均为低度多态(PIC<0.25),CLEC16A基因g.9587011 G>A、g.9730880 G>A位点在3个群体中为中度多态(0.25<PIC<0.5);卡方检验结果显示,ANKRD17基因g.88739493 T>G位点以及CLEC16A基因g.9587011 G>A、g.9730880 G>A位点在3个山羊品种中均处于Hardy-Weinberg不平衡状态(P<0.05);与繁殖性能相关的表型关联分析表明,CLEC16A基因g.9587011 G>A位点中GA基因型的个体产羔数显著高于GG基因型个体(P<0.05),3个SNPs位点对初生窝重和断奶窝重均无显著影响(P>0.05)。综上,CLEC16A基因g.9587011 G>A位点可作为山羊产羔数选择的潜在分子标记。
The purpose of this study was to explore the polymorphisms of ANKRD17(g.88739493 T>G)gene and CLEC16A(g.9587011 G>A and g.9730880 G>A)gene and their association with reproductive performance in goats.Sequenom MassARRAY^(■)SNP typing technology was used to detect the polymorphisms of ANKRD17 and CLEC16A genes in Yunshang Black goat,Jining Gray goat and Liaoning cashmere goat,then the association between ANKRD17 and CLEC16A and reproductive performance(litter size,litter weight at birth and litter weight at weaning)in goats was analyzed.The results showed that ANKRD17 gene g.88739493 T>G had low polymorphism(PIC<0.25)in the three goats;CLEC16A gene g.9587011 G>A and g.9730880 G>A were moderately polymorphic in the three populations(0.25<PIC<0.5).The Chi-square test results showed that ANKRD17 gene g.88739493 T>G locus and CLEC16A gene g.9587011 G>A and g.9730880 G>A locus were under Hardy-Weinberg disequilibrium in the three goats(P<0.05).The association analysis of genotypes related to reproductive performance showed that there was a significant correlation between the polymorphism of CLEC16A gene g.9587011 G>A locus and the litter size of Yunshang Black goat,the litter size of GA was higher than GG(P<0.05),while there was no significant correlation between the three SNPs locus and the litter weight at birth and litter weight at weaning in all goats(P>0.05).In summary,the g.9587011 G>A locus at CLEC16A gene can be used as a potential molecular marker for litter size in goats.
