八宝眼膏对暴露性角膜炎大鼠治疗作用的研究

Study on the therapeutic effects of BaBao eye ointment on the experimental exposure keratitis rat model

目的观察八宝眼膏对暴露性角膜炎大鼠的治疗作用。方法将32只大鼠随机分为4组,每组8只(8只眼):正常组、模型组、治疗组(八宝眼膏组)、阳性对照组(维生素A棕榈酸酯眼用凝胶组)。将除正常组以外,其他组采用上下眼睑缝线固定法制造暴露性角膜炎模型,阳性对照组和治疗组分别于6 d后给予维生素A棕榈酸酯眼用凝胶和八宝眼膏涂眼,每日2次。正常组和模型组不予处理。3 d后,对各组大鼠进行眼表检查,检测泪液分泌量(SⅠt),荧光素钠染色(FL)、泪膜破裂时间(BUT)、组织病理学、Muc-5ac表达等。结果(1)FL:正常组、阳性对照组和治疗组大鼠的FL为阴性。模型组大鼠FL为强阳性;(2)SⅠt:正常组与模型组相比,模型组大鼠的SⅠt显著降低(t=4.686,P=0.000)。与模型组比较,阳性对照组、治疗组均显著增高(t_(阳性)=4.246,t_(治疗)=4.393;均P=0.001);(3)BUT:模型组BUT较正常组显著降低(t=13.267,P=0.000),与模型组相比,阳性对照组和治疗组均显著增高(t_(阳性)=12.507,t_(治疗)=14.877;均P=0.000);(4)组织病理学:与模型组相比,治疗组、阳性对照组的角膜组织结构的完整性都有所改善,上皮层细胞排列较为紧密,实质层细胞的形态也较为规则;(5)Muc-5ac表达:与模型组相比,正常组、治疗组、阳性对照组Muc-5ac阳性着色部位均呈现较深颜色。结论八宝眼膏对暴露性角膜炎引起的泪膜稳定性下降,角膜和结膜的损伤均有治疗作用。

OBJECTIVE To observe the therapeutic effects of BaBao eye ointment on exposure keratitis.METHODS Thirty-two rats were randomly divided into 4 groups(8 eyes per group):normal control group,model group,treatment group(BaBao eye ointment group,TG),positive control group(vitamin A palmitate eye gel group,PG).Experimental exposure keratitis rat model was made by upper and lower eyelid suture fixation in all groups except for the normal group.The positive control group and the treatment group were given vitamin A palmitate ophthalmic gel and Babao eye ointment twice per day on day 6 after model establishment,respectively.No treatment was givin to the normal group and the model group.Three days later,eye surface examination was performed to detect Schirmer I test(SⅠt),fluorescein vital staining(FL),tear break-up time(BUT),and histopathological expression of Muc-5ac.RESULTS(1)FL:The corneal fluorescein vital staining was all negative in the normal group,the positive control group and the Babao eye ointment group.Fluorescence staining of the cornea and conjunctiva was strongly positive in the model group.(2)SⅠt:Compared with the normal group,the tear secretion of rats in the model group was significantly reduced(t=4.686,P=0.000),while that in the positive control group and the Babao eye ointment group was significantly higher when compared to the model group(t_(PG)=4.246,t_(TG)=4.393,all P=0.001).(3)BUT:The tear film rupture time in the model group was significantly lower than that in the normal group(t=13.267,P=0.000),while that in the positive control group and the Babao eye ointment group was significantly higher compared to the model group(t_(PG)=12.507,t_(TG)=14.877,all P=0.000).(4)Histopathology:When compared to the model group,the integrity of corneal tissue structure in the Babao eye ointment group and the vitamin A palmitate eye gel group was both improved,with closer arrangements of cells in the upper cortex and more regular cell morphology in the parenchymal layer.(5)Muc-5ac expression:Compared with the model group,the positive staining sites of Muc-5ac demonstrated darker color in the normal group,the Babao eye ointment group and the vitamin A palmitate eye gel group.CONCLUSIONS BaBao eye ointment has therapeutic effects on decreased tear film stability as well as corneal and conjunctival injuries cause by exposure conjunctivitis.
