高碘对NOD小鼠甲状腺组织c-fos表达的影响

摘要 目的研究高碘对NOD小鼠甲状腺组织c-fos表达的影响。方法选取5周龄的雌性NOD小鼠20只,随机分为对照组(NC)、10倍高碘组(10 HI)、100倍高碘组(100 HI)、1000倍高碘组(1000 HI),每组5只。NC组饮用去离子水,10 HI组饮用含0.0006%碘化钠去离子水,100 HI组饮用含0.006%碘化钠去离子水,1000 HI组饮用含0.06%碘化钠去离子水,无碘饲料喂养。在第16周结束时处死小鼠。实时荧光定量核酸扩增检测系统检测甲状腺组织c-fos mRNA表达水平,蛋白免疫印迹法(Western blot)检测甲状腺组织c-fos蛋白表达水平。结果4组小鼠甲状腺组织c-fos mRNA水平组间比较有显著差异(P<0.05),且随着碘摄入量的增高甲状腺组织c-fos mRNA表达水平显著增高。蛋白表达组间有显著统计学差异(P<0.05)。结论高浓度碘的摄入可促进NOD小鼠甲状腺组织c-fos表达。 Objective To study the effect of high iodine on c-fos expression in thyroid tissue of NOD mice.Methods Twenty 5-week-old female NOD mice were selected and randomly divided into control group(NC),10-fold high iodine group(10 HI),100-fold high phosphorus group(100 HI),and 1000-fold high NOD group(1000 HI),with 5 mice in each group.The NC group was fed with deionized water,the 10 HI group was fed with deionized water containing 0.0006%iodine,the 100 HI group was fed with deionized water containing 0.006%iodine,and the 1000 HI group was fed with deionized water containing 0.06%iodine.The mice were killed at the end of week 16.The mRNA expression level of c-fos in thyroid tissues was detected by real-time PCR,and the protein expression level of c-fos in thyroid tissues was detected by Western blot.Results The mRNA level of c-fos in thyroid tissue of mice was significantly different among the four groups(P<0.05),and the expression level of c-fos mRNA in thyroid tissue was significantly increased with the increase of iodine intake.There was statistically significant difference in protein expression between groups(P<0.05).Conclusion High concentration of iodine intake can promote the expression of c-fos in thyroid tissue of NOD mice.
