红毛藻多糖对H_(2)O_(2)诱导的Caco-2细胞氧化损伤的保护作用 被引量:8

Protective Effect of Bangia fusco-purpurea Polysaccharide on H_(2)O_(2)-Induced Oxidative Damage in Caco-2 Cells

摘要 目的:研究红毛藻多糖(Bangia fusco-purpurea polysaccharide,BFP)对H_(2)O_(2)诱导的人结肠腺癌(Caco-2)细胞氧化应激损伤的保护作用。方法:构建过氧化氢(H_(2)O_(2))诱导的Caco-2细胞氧化损伤模型,通过形态学观察和测定细胞裂解液中的抗氧化物水平及相关酶活力,评价BFP对Caco-2细胞氧化损伤的保护作用。结果:BFP可明显提高H_(2)O_(2)诱导氧化损伤的Caco-2细胞活力,降低细胞内活性氧生成水平,提高细胞内超氧化物歧化酶和过氧化氢酶活力以及还原型谷胱甘肽水平,减少丙二醛生成,并通过抑制Caspase-3和Caspase-9活性来改善氧化应激损伤引起的细胞凋亡。结论:BFP对H2O2诱导氧化应激损伤的Caco-2细胞具有保护作用,可通过增强细胞内源性抗氧化酶活力、提高非酶类抗氧化物水平和抑制细胞凋亡明显缓解H_(2)O_(2)诱导的Caco-2细胞氧化应激损伤。 Objective:The protective effect of Bangia fusco-purpurea polysaccharide(BFP)on H_(2)O_(2)-induced oxidative damage in human clone colon cancer cells(Caco-2 cells)was investigated in this study.Methods:The morphology of the oxidatively damaged cells was observed and antioxidant compound contents and antioxidant enzyme activities in the cell lysates were measured.Results:BFP significantly increased the viability of Caco-2 cells damaged by H_(2)O_(2) in a concentration-dependent manner,reduced the generation of intracellular reactive oxygen species(ROS),increased the activity of superoxide dismutase(SOD)and catalase(CAT)and glutathione level,inhibited the production of malondialdehyde(MDA),and repressed caspase-3 and caspase-9 activity to mitigate cell apoptosis induced by oxidative stress.Conclusion:BFP has a protective effect on H_(2)O_(2)-induced oxidative damage in Caco-2 cells in vitro.It can ameliorate H_(2)O_(2)-induced oxidative stress damage in Caco-2 cells by enhancing the activities of endogenous antioxidant enzymes,increasing the levels of non-enzymatic antioxidants,and inhibiting cell apoptosis.
