摘要
目的:运用网络药理学研究自拟桑蚕颗粒治疗2型糖尿病的作用机制,为新药研发提供支撑。方法:通过文献挖掘和数据库检索得到桑蚕颗粒活性成分及潜在靶点;在GEO数据库中筛选2型糖尿病基因芯片,经差异分析获取差异基因作为疾病靶点。应用Cytoscape3.7.2软件构建中药复方调控网络图。利用Cytoscape3.7.2插件Bisogenet构建蛋白质-蛋白质相互作用网络(PPI),CytoNCA插件对其进行拓扑分析,MCODE插件对PPI核心网络进行功能聚类,应用Bioconductor分别对各功能模块实现GO功能富集分析和KEGG通路富集分析。结果:研究得到97个活性成分和42个关键靶点,PPI核心网络主要有4个功能模块,GO功能富集分析中生物学过程共涉及1396个条目,细胞组分共涉及113个条目,分子功能共涉及153个条目,KEGG富集分析共涉及17条通路。结论:自拟桑蚕颗粒有效成分可调控2型糖尿病发病重要通路中多个靶点。
Objective:To study the mechanism of action of self-made Sangcan Granules(自拟桑蚕颗粒)in the treatment of typeⅡdiabetes by network pharmacology,and to provide support for the development of new medicines.Methods:Through literature mining and database search,the active ingredients and potential targets of Sangcan Granules were obtained.The gene chip of typeⅡdiabetes was screened in GEO database,and differential genes were obtained as disease targets by differential analysis.Cytoscape3.7.2 software was used to construct a network regulation and control diagram of Chinese herbal compound.Cytoscape3.7.2 plug-in Bisogenet was used to construct protein-protein interaction network(PPI).CytoNCA plug-in was used for topology analysis.MCODE plug-in was used for functional clustering of PPI core network,and Bioconductor was used to implement GO function enrichment analysis and KEGG pathway enrichment analysis for each functional module.Results:This study obtained total 97 active ingredients and 42 key targets.The PPI core network mainly has 4 functional modules.In GO functional enrichment analysis,the biological process involves a total of 1396 items,and the cell component involves a total of 113 items.The molecule function involves a total of 153 items,and the KEGG enrichment analysis involves a total of 17 pathways.Conclusions:The active ingredients of self-made Sangcan Granules can regulate many targets in the important pathways of typeⅡdiabetes.
