丙泊酚对感染性休克大鼠肾功能和PI3K/Akt信号通路的影响 被引量:1

Effects of propofol on renal function and PI3K/Akt signaling pathway in septic shock rats model

摘要 目的探讨丙泊酚对感染性休克大鼠肾功能和PI3K/Akt信号通路的影响。方法将SPF级Wistar大鼠分为对照组、模型组、丙泊酚组、LY294002P组、丙泊酚+LY294002P组,检测各组大鼠尿素氮(BUN)、血肌酐(Scr)、半胱氨酸抑制素C(CysC)、乳酸脱氢酶(LDH)、超氧化物岐化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)。结果与对照组相比,模型组、丙泊酚组、LY294002P组、丙泊酚+LY294002P组的Scr、BUN、CysC、LDH、MDA、TNF-α、IL-6、IL-1β、肾组织凋亡指数、Cleaved-Cas-3、Bax蛋白水平升高(P<0.05),SOD、GSH-Px含量和Bcl-2、p-PI3K/PI3K、pAKT/AKT蛋白水平降低(P<0.05)。与模型组相比,丙泊酚组、丙泊酚+LY294002P组Scr、BUN、CysC、LDH、MDA、TNF-α、IL-6、IL-1β、肾组织凋亡指数、Cleaved-Cas-3、Bax蛋白水平降低(P<0.05),SOD、GSH-Px含量和Bcl-2、p-PI3K/PI3K、pAKT/AKT蛋白水平升高(P<0.05);与模型组相比,LY294002P组Scr、BUN、CysC、LDH、MDA、TNF-α、IL-6、IL-1β、肾组织凋亡指数、Cleaved-Cas-3、Bax蛋白水平升高(P<0.05),SOD、GSH-Px含量和Bcl-2、p-PI3K/PI3K、pAKT/AKT蛋白水平降低(P<0.05)。与丙泊酚组相比,丙泊酚+LY294002P组Scr、BUN、CysC、LDH、MDA、TNF-α、IL-6、IL-1β、肾组织凋亡指数、Cleaved-Cas-3、Bax水平升高(P<0.05),SOD、GSH-Px含量和Bcl-2、p-PI3K/PI3K、pAKT/AKT蛋白水平降低(P<0.05)。结论丙泊酚可以改善感染性休克大鼠肾功能损伤,可能与降低炎症和氧化应激反应、激活PI3K/Akt信号通路有关。 OBJECTIVE To explore the effects of propofol on renal function and PI3 K/Akt signaling pathway in septic shock rats model.METHODS SPF Wistar rats were divided into the control group,model group,propofol group,LY294002 P group and propofol+LY294002 P group,in which urea nitrogen(BUN),blood creatinine(Scr)and cysteine Inhibin C(CysC),lactate dehydrogenase(LDH),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),malondialdehyde(MDA),tumor necrosis factor-α(TNF-α),Interleukin-6(IL-6)and Interleukin-1β(IL-1β)were detected.RESULTS Compared with the control group,Scr,BUN,CysC,LDH,MDA,TNF-α,IL-6,IL-1β,apoptosis index of renal tissue,Cleaved-Cas-3 and Bax protein levels in the model group,propofol group,LY294002 P group and propofol+LY294002 P group were significantly increased(P<0.05),while SOD,GSH-Px content,Bcl-2,p-PI3 K/PI3 K and pAKT/AKT proteins levels were significantly decreased(P<0.05).Compared with the model group,Scr,BUN,CysC,LDH,MDA,TNF-α,IL-6,IL-1β,apoptosis index of renal tissue,cleaved-Cas-3 and Bax protein levels in the propofol group and propofol+LY294002 P group were significantly decreased(P<0.05),while SOD,GSH-Px content,Bcl-2,p-PI3 K/PI3 K and pAKT/AKT proteins levels were significantly increased(P<0.05).Compared with model group,Scr,BUN,CysC,LDH,MDA,TNF-α,IL-6,IL-1β,apoptosis index of renal tissue,Cleaved-Cas-3 and Bax protein levels in the LY294002 P group were significantly increased(P<0.05),while SOD,GSH-Px content,Bcl-2,p-PI3 K/PI3 K and pAKT/AKT proteins levels were significantly decreased(P<0.05).Compared with the propofol group,Scr,BUN,CysC,LDH,MDA,TNF-α,IL-6,IL-1β,apoptosis index of renal tissue,cleaved-Cas-3 and Bax protein levels were significantly increased in the propofol+LY294002 P group(P<0.05),while SOD,GSH-Px content,Bcl-2,p-PI3 K/PI3 K and pAKT/AKT proteins levels were significantly decreased(P<0.05).CONCLUSION Propofol can alleviate renal function injury in septic shock rats,which may be related to reducing inflammation and oxidative stress response,and activating PI3 K/Akt signaling pathway.
