芝麻酚对模型小鼠肝损伤的改善作用 被引量:1

摘要 目的探讨芝麻酚对模型小鼠肝损伤的改善作用。方法将KM小鼠分为正常对照组(等体积0.2%CMC-Na溶液)、模型组(等体积0.2%CMC-Na溶液)、水飞蓟素组(200mg/kg),以及芝麻酚低、中、高剂量组(6,12,24mg/kg)。正常对照组小鼠仅灌胃等体积0.2%CMC-Na溶液,其余各组小鼠灌胃相应药物及黄药子75%乙醇提取物(18g/kg),每日1次,持续30d。检测血浆中生化指标天冬氨酸氨基转移酶(AST)、丙氨酸氨基转氨酶(ALT)、碱性磷酸酶(ALP)、总胆红素(TBiL)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、还原型谷胱甘肽(GSH)、丙二醛(MDA)水平,并对小鼠肝脏组织进行病理切片检查。采用Western blotting法检测肝组织匀浆中肿瘤坏死因子-α(TnF-α)、白细胞介素6(IL-6)、核因子κB(NF-κB)p65、核因子E2相关因子2(Nrf2)、血红素加氧酶(HO-1)的水平。结果与模型组比较,芝麻酚低、中、高剂量组的ALT,AST,TBiL,MDA及NF-κBp65,TNF-α,IL-6蛋白表达水平均显著降低,GSH及HO-1蛋白表达水平显著升高(P<0.05或P<0.01);芝麻酚高剂量组的SOD和CAT水平均显著升高(P<0.05或P<0.01)。模型组小鼠肝细胞肿胀,边缘模糊,出现细胞核溶解,芝麻酚低、中、高剂量组肝细胞明显改善,损伤较少。结论芝麻酚可改善黄药子75%乙醇提取物诱导的小鼠肝损伤,其机制可能与调节Nrf2/NF-κB信号通路而提高机体的抗氧化和抗炎能力有关。 Objective To investigate the effect of sesamol on the improvement of liver injury in model mice.Methods KM mice were divided into the normal control group(equal volume of 0.2%CMC-Na solution),model group(equal volume of 0.2%CMC-Na solution),silymarin group(200 mg/kg)and sesamol low-dose,medium-dose and high-dose groups(6,12,24 mg/kg).Mice in the normal control group were only gavaged with an equal volume of 0.2%CMC-Na solution,and mice in other groups were gavaged with corresponding drugs and 75%ethanol extract(18 g/kg)of Rhizoma Dioscoreae Bulbiferae once a day for 30 d.The levels of aspartate aminotransferase(AST),alanine aminotransferase(ALT),alkaline phosphatase(ALP),total bilirubin(TBiL),superoxide dismutase(SOD),catalase(CAT),reduced glutathione(GSH)and malondialdehyde(MDA)in plasma were measured.The liver tissue of mice was examined by the pathological section.The levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),nuclear factor-κB(NF-κB)p65,nuclear factor E2-related factor 2(Nrf2)and heme oxygenase-1(HO-1)in the liver homogenate was detected by Western blotting.Results Compared with those in the model group,the levels of ALT,AST,TBiL,MDA and NF-κBp65,TNF-αand IL-6 decreased significantly,while the expression levels of GSH and HO-1 protein increased significantly in the low-dose,medium-dose and high-dose sesamol groups(P<0.05 or P<0.01),the levels of SOD and CAT in the high-dose sesamol group increased significantly(P<0.05 or P<0.01).The hepatocytes in the model group were swollen,the edges were blurred,and the nuclei were dissolved.The hepatocytes of mice in the low-dose,medium-dose and high-dose sesamol groups were significantly improved with less damage.Conclusion Sesamol can improve the liver injury induced by 75%ethanol extract of Rhizoma Dioscoreae Bulbiferae,and its mechanism may be related to regulating Nrf2/NF-κB signal pathway,so as to improve the antioxidant and anti-inflammatory ability of the body.
