“智三针”对Shank3慢病毒干扰的孤独症模型鼠行为学影响 被引量:3

摘要 目的观察“智三针”对Shank3孤独症模型鼠行为学的影响。方法通过Shank3慢病毒干扰Wistar孕鼠,出生后仔鼠随机分为模型组、针刺组及假针刺组;采用空载慢病毒干扰Wistar孕鼠,出生后仔鼠为对照组,每组10只。针刺组采用“智三针”干预,假针刺组采用非经非穴针刺干预。通过体质量、负向趋地性反应、游泳实验观察Shank3慢病毒干扰对各组仔鼠体格生长、前庭平衡功能、感觉机能及运动协调能力发育的差异;通过旷场、Morris水迷宫及三箱社交实验,观察“智三针”对各组仔鼠自主探索活动、空间学习记忆和社会交往能力等行为学的影响。结果与对照组比较,Shank3模型鼠体格生长、前庭平衡功能、感觉机能及运动协调能力发育无明显变化。与对照组比较,模型组在旷场实验中的运动总距离、中间区域的逗留时间及探索次数均减少(P<0.01,P<0.05);水迷宫实验中目标象限时间、游泳距离和平台穿越次数均减少(P<0.01);三箱实验中探索物品或互动社交伙伴的时间无明显变化(P>0.05),对新旧动物的社交偏爱并未表现出差异(P>0.05),接触新动物的时间减少(P<0.05)。与模型组和假电针组比较,针刺组在旷场中的运动总距离和中间区域的逗留时间增加(P<0.05),探索次数差异无统计学意义(P>0.05);水迷宫中目标象限时间和平台穿越次数均增加(P<0.05),游泳距离差异无统计学意义(P>0.05);在三箱实验中互动社交伙伴的时间多于探索物品(P<0.05),对新旧动物的社交偏爱并未表现出差异(P>0.05),接触新动物的时间增多(P<0.05)。结论“智三针”可改善Shank3孤独症模型鼠的自主活动、空间学习记忆和社会交往能力,而对新事物的喜好行为的作用仍需进一步研究。 Objective To observe the effects of Zhi San Zhen(acupuncture at three acupoints specifically for improving the intelligence)on the behaviors of Shank3 autism model rats.Method The newborns of pregnant Wistar rats infected by Shank3 lentivirus were randomized into a model group,an acupuncture group and a sham acupuncture group,and a control group was set with the newborns of those infected by an empty vector,with 10 rats in each group.The acupuncture group was intervened using Zhi San Zhen and the sham acupuncture group was intervened using non-acupoint spots.Body weight,negative geotactic response and swimming experiment were used to observe the influences of Shank3 lentivirus interference on the physical growth,vestibular balance function,sensory function and motor coordination ability of the rats;open field test,Morris water maze and three chamber free exploring test were conducted to observe the effects of Zhi San Zhen on the behaviors of rats,including autonomous exploration activities,spatial learning and memory,and social interaction ability.Result Compared with the control group,there were no significant changes in the development of physical growth,vestibular balance function,sensory function and motor coordination in Shank3 model rats.Compared with the control group,the total distance,stay in the center area and the number of explorations in the open field test were reduced in the model group(P<0.01,P<0.05);the stay in the target quadrant,swimming distance and number of platform crossing in the water maze experiment were reduced(P<0.01);there was no significant difference in the time of exploring objects or interaction(P>0.05),nor in the social preference for old or new animals(P>0.05);the time of exposure to new animals decreased(P<0.05).Compared with the model group and sham acupuncture group,the acupuncture group had increased total distance and stay in the center area in the open field test(P<0.05),but without significant difference in the number of explorations(P>0.05);the stay in the target quadrant and number of platform crossing in the water maze test also increased(P<0.05),while the swimming distance was not significantly different(P>0.05);in the three chamber experiment,more time was spent on interacting with companions rather than exploring objects(P<0.05),while there was no significant difference in the social preference for old or new animals(P>0.05),and more time was spent on exposure to new animals(P<0.05).Conclusion Zhi San Zhen can improve the autonomous activities,spatial learning and memory and social interaction abilities in the Shank3 autism rats,while its effect on the preference for new things requires further study.
