大孔树脂分离纯化山慈菇总黄酮的工艺研究 被引量:1

Separation and Purification of Total Flavonoids from Shancigu(Cremastrae Pseudobulbus Pleiones Pseudobulbus) by Macroporous Resin

摘要 目的探讨山慈菇总黄酮的大孔树脂分离纯化工艺。方法比较11种大孔树脂对山慈菇总黄酮的静态吸附率和解析率,筛选出最佳树脂。通过单因素实验,优化最佳树脂动态吸附与解析总黄酮的工艺参数。结果 HPD-750型大孔树脂对山慈菇中总黄酮有较好的吸附与洗脱效果。最佳工艺为:上样液质量浓度173.35μg·mL^(-1),体积流量1 mL·min^(-1),pH为5,洗脱液(乙醇)体积流量1.5 mL·min^(-1),体积分数70%。在该工艺条件下,山慈菇中总黄酮的质量分数为44.30%,回收率为75.70%。结论 HPD-750树脂综合性能良好,适用于山慈菇中总黄酮的分离纯化。 Objective To study the separation and purification of total flavonoids from Shancigu (Cremastrae Pseudobulbus Pleiones Pseudobulbus) by macroporous resin.Methods Comparing the static adsorption rate and resolution rate of total flavonoids of 11 macroporous resins,the best resin was screened out.The parameters of the optimal resin dynamic adsorption and desorption total flavonoid were optimized by single factor experiment.Results HPD-750 macroporous resin was found to have good adsorption and desorption effects.The optimal conditions were as follows.The mass concentration of the sample solution was 173.35μg·m L-1.The volume flow rate was 1 mL·min^(-1).The p H value was 5.The volume flow rate of eluent(ethanol)was 1.5 mL·min^(-1).The volume fraction was 70%.Under the conditions of the process,the content of total flavonoids in Shancigu(Cremastrae Pseudobulbus Pleiones Pseudobulbus) was 44.30%,and the recovery rate was 75.70%.Conclusion HPD-750 resin has good comprehensive performance and is suitable for the separation and purification of total flavonoids from Shancigu(Cremastrae Pseudobulbus Pleiones Pseudobulbus).
