胃癌患者血清中miRNA-181s表达与幽门螺杆菌感染的关系

Serum level of miRNA-181s in gastric cancer patients and its correlation with Helicobacter pylori infection

目的探讨胃癌患者血清中微小RNA(miRNA)-181s表达水平,及其与幽门螺杆菌(Hp)感染的关系。方法用尿素14 C呼气试验检测该院2019年1月至2020年6月收治的50例胃癌患者的Hp感染情况。采用实时荧光定量PCR检测胃癌患者(观察组)和体检健康者(对照组)血清miRNA-181s相对表达水平,进一步分析miRNA-181s相对表达水平与Hp感染的关系。采用酶联免疫吸附试验法检测白细胞介素(IL)-6和磷酸化信号传导子及转录激活子3(p-STAT3)表达水平。结果观察组Hp感染率明显高于对照组,差异有统计学意义(P<0.05)。观察组Hp阳性患者miRNA-181s相对表达水平均明显高于Hp阴性患者,差异有统计学意义(t 1=7.449、t 2=8.828、t 3=6.478、t 4=6.060,P<0.05)。观察组Hp阳性患者和Hp阴性患者血清中IL-6表达水平比较,差异有统计学意义(P<0.05);观察组Hp阳性患者和Hp阴性患者血清中p-STAT3表达水平比较,差异有统计学意义(P<0.05)。结论胃癌患者血清中miRNA-181a、miRNA-181b、miRNA-181c和miRNA-181d表达水平与Hp感染有关,并且IL-6/p-STAT3信号途径可能参与胃癌患者Hp感染过程。

Objective To investigate the expression level of microRNA(miRNA)-181s in serum of gastric cancer patients and its relationship with Helicobacter pylori(Hp)infection.Methods Hp infection was detected in 50 patients with gastric cancer in our hospital from January 2019 to June 2020 by the 14 C-urea breath test.The relative expression level of miRNA-181s in serum of gastric cancer patients(observation group)and healthy subjects(control group)was detected by real-time fluorescence quantitative PCR,and the relationship between the relative expression level of miRNA-181s and Hp infection was further analyzed.Enzyme-linked immunosorbent assay was used to determine the secretion of IL-6 and p-STAT3.Results The infection rate of Hp in the observation group was significantly higher than that in the control group,and the difference was statistically significant(P<0.05).The relative expression level of miRNA-181s in Hp positive patients was significantly higher than that in Hp negative patients,with statistical significance(t 1=7.449,t 2=8.828,t 3=6.478,t 4=6.060,P<0.05).The expression level of IL-6 in serum of Hp-positive patients and Hp-negative patients in observation group was statistically significant(P<0.05).The expression level of p-STAT3 in serum of Hp-positive patients and Hp-negative patients in observation group was statistically significant(P<0.05).Conclusion The expression levels of miRNA-181a,miRNA-181b,miRNA-181c and miRNA-181d in serum of gastric cancer patients are related to Hp infection,and the IL-6/p-STAT3 signaling pathway may be involved in this process.
