正交实验法优化盐地碱蓬中多糖的超声提取工艺 被引量:1

摘要 目的:正交实验法优化盐地碱蓬中多糖的提取工艺。方法:利用正交实验对盐地碱蓬中多糖的提取工艺进行优化。以盐地碱蓬中多糖的提取率为技术指标,采用超声提取方法,选取提取的温度、料液比、提取的次数、提取的时间为影响因素,在单因素试验的基础上,对超声提取工艺利用正交试验进行优化,采用苯酚-硫酸法测定多糖的含量。结果:根据单因素试验的结果,开展正交试验,得到最理想的盐地碱蓬多糖的超声提取工艺是料液比1∶40,超声提取的次数3次,超声提取的时间30 min,超声提取的温度40℃。结论:优化的超声提取盐地碱蓬多糖工艺操作简便易行,稳定性好,具有实用价值,可依据该工艺条件制定盐地碱蓬多糖提取工艺。 Objective: To optimize the extraction technology of polysaccharide from Suaeda salsa by orthogonal experiment. Method: Orthogonal experiment was used to optimize the extraction process of polysaccharide from Suaeda salsa by using ultrasonic extraction. To salt alkaline extraction yield of polysaccharide as index, refluxing extraction selected material liquid ratio, extraction temperature, extraction time, extraction times for the influence factors, on the basis of single factor experiment, orthogonal test, process optimization, using the phenol – sulfuric acid method for determining the content of polysaccharide. Results: Based on the results of the single factor experiments and orthogonal experiments, the extraction process of Suaeda salsa polysaccharide was as follows: solid-liquid ratio of 1∶30,times of 3, time of 30 min, temperature of 40 ℃. Conclusion: The extraction methods of Suaeda salsa polysaccharide is simple, easy to operate, good stability, and has practical value. The extraction process of Suaeda salsa polysaccharide can be developed according to the technological conditions.
