摘要 目的:建立一测多评法同时测定金花清感颗粒中黄芩苷、绿原酸、芒果苷、汉黄芩苷和甘草酸铵的含量。方法:采用高效液相色谱法,Agilent Eclipse XDB-C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.1%磷酸水溶液为流动相进行梯度洗脱,流速1.0 ml/min,检测波长327、254和277 nm,柱温25℃,进样量10μl。以黄芩苷为内参物,计算其他5个成分的相对校正因子,并计算各成分含量。结果:绿原酸、芒果苷、黄芩苷、牛蒡苷、汉黄芩苷和甘草酸铵的线性范围分别为4.98~49.85、2.05~20.52、22.84~228.40、41.02~410.22、7.40~74.03和4.93~49.34μg/ml(r﹥0.999)。平均加样回收率分别为98.00%、101.07%、97.69%、96.92%、96.79%和102.11%,RSD值分别为1.53%、1.78%、1.37%、1.62%、1.68%和1.67%。在上述线性范围内,黄芩苷与绿原酸、芒果苷、牛蒡苷、汉黄芩苷、甘草酸铵的相对校正因子分别为1.106、0.818、6.065、0.860和4.635。一测多评法所测结果与外标法无明显差异。结论:该法简便、重复性好,可为金花清感颗粒的质量评价提供参考。 Objective:To establish a quantitative analysis of multi-components by single-marker(QAMS)method for the simultaneous determination of baicalin,chlorogenic acid,mangiferin,arctiin,wogonoside,and ammonium glycyrrhizinate in Jinhua Qinggan granules.Methods:HPLC was performed on Agilent Eclipse XDB-C18(250 mm×4.6 mm,5μm)column with acetonitrile-0.1%phosphoric acid solution for gradient elution.The flow rate was 1.0 ml/min,and the detection wavelengths were 327,254,and 277 nm.Baicalin was used as an internal standard to calculate the relative correction factors of the other five components,and the contents of each component were calculated.Results:The linear ranges of chlorogenic acid,mangiferin,baicalin,arctiin,wogonoside,and ammonium glycyrrhizinate were 4.98~49.85,2.05~20.52,22.84~228.40,41.02~410.22,7.40~74.03 and 4.93~49.34μg/ml,respectively(r﹥0.999).The average recoveries were 98.00%,101.07%,97.69%,96.92%,96.79%and 102.11%and RSD values were 1.53%,1.78%,1.37%,1.62%,1.68%and 1.67%,respectively.Within the above linear range,the relative correction factors of chlorogenic acid,mangiferin,arctiin,wogonoside,and ammonium glycyrrhizinate to baicalin were 1.106,0.818,6.065,0.860 and 4.635,respectively.There was no significant difference between the calculated values by QAMS and the measured values by the external standard method.Conclusion:The method is simple and reproducible,which can provide a reference for the quality evaluation of Jinhua Qinggan granules.
