灵芝三萜通过Wnt信号通路对胶质瘤细胞侵袭和迁移的影响 被引量:1

摘要 目的分析灵芝三萜通过Wnt信号通路对胶质瘤细胞侵袭和迁移的影响。方法用10 mg/L(低剂量组)、50 mg/L(中剂量组)、100 mg/L(高剂量组)的灵芝三萜处理人胶质瘤U251细胞,并设置对照组(未给予灵芝三萜处理);比较处理不同时刻及不同浓度灵芝三萜对胶质瘤细胞U251的增殖、侵袭、迁移及凋亡的影响,同时检测U251细胞中β连环蛋白(β-catenin)、磷酸化糖原合成酶激酶3β(p-GSK-3β)蛋白的表达水平。结果药物处理不同时刻对照组U251细胞的OD值、侵袭细胞数、迁移率、凋亡率、β-catenin、p-GSK-3β蛋白表达均高于3个灵芝三萜组,差异有统计学意义(P<0.05);随着处理灵芝三萜浓度的升高,U251细胞的OD值、侵袭细胞数、迁移率、凋亡率均逐渐降低,β-catenin、p-GSK-3β蛋白表达均逐渐升高,差异有统计学意义(P<0.05);随着处理时间的延长转染同浓度的灵芝三萜U251细胞的OD值、侵袭细胞数、迁移率、凋亡率均升高,β-catenin、p-GSK-3β蛋白表达均降低,差异有统计学意义(P<0.05)。结论灵芝三萜可能够通过Wnt信号通路抑制胶质瘤细胞的增殖、侵袭和迁移,促进细胞凋亡。 Objective To analyze the effect of triterpenes of ganoderma lucidum(G.Lucidum)on invasion and migration of glioma cells through Wnt pathway.Methods Human glioma U251 cells were transfected with triterpenes of G.Lucidum at 10 mg/L(low-dose group),50 mg/L(medium-dose group),100 mg/L(high-dose group),negative control(negative control group)and untreated(blank control group).The effects of triterpenoids of G.lucidum at different concentrations and different time on cell proliferation,invasion,migration,apoptosis were examined.Additionally,the protein levels ofβ-catenin and phosphorylation-glycogen synthase kinase-3β(p-GSK-3β)were detected in glioma cells.Results The OD values at 490 nm,the number of invaded cells and the migration rates,apoptotic rates,the protein levels ofβ-catenin and p-GSK-3βwere higher in blank control and the negative control groups than those of triterpenoids of G.lucidum-transfected cells(P<0.05).OD values,number of invaded cells,migration rate,and apoptosis rate of U251 cells gradually decreased in a dose of G.Lucidum-dependent manner,the protein levels ofβ-catenin and p-GSK-3βwere increased(P<0.05).In contrast,OD values,number of invaded cells,migration rate,and apoptosis rate of U251 cells gradually increased in a time-dependent manner,the protein levels ofβ-catenin and p-GSK-3βwere decreased(P<0.05).Conclusion Triterpenes of G.Lucidum can inhibit cell proliferation,invasion and migration,and promote cell apoptosis of glioma cells through Wnt signaling pathway.
