结肠癌组织中miR-129-5p、BDNF表达变化及意义 被引量:4

Expression and significance of miR-129-5p and BDNF in colon cancer tissues

摘要 目的探讨结肠癌组织中微小RNA-129-5p(miR-129-5p)、脑源性神经营养因子(BDNF)的表达变化及临床意义。方法选取118例结肠癌患者,均接受根治性或姑息性手术,取术中切除的结肠癌组织及癌旁组织(距离肿瘤组织边缘≥5 cm),采用qRT-PCR检测组织中miR-129-5p、BDNF mRNA表达,用Spearman相关法分析二者的相关性,分析二者表达变化与结肠癌患者临床病理特征的关系;对患者进行随访,统计3年总生存率,分析miR-129-5p、BDNF mRNA表达变化与患者预后的关系。结果与癌旁组织比较,结肠癌组织中miR-129-5p相对表达量低,BDNF相对表达量高(P均<0.05)。miR-129-5p、BDNF mRNA表达与结肠癌分化程度、TNM分期、浸润深度、淋巴结转移有关(P均<0.05),与患者性别、年龄、肿瘤直径无关(P均>0.05)。结肠癌组织中miR-129-5p表达与BDNF mRNA表达呈负相关(r=-5.174,P<0.05)。以结肠癌组织中miR-129-5p、BDNF mRNA表达的中位数为基准,将结肠癌患者分为高、低表达组。miR-129-5p高表达组3年总生存率高于miR-129-5p低表达组,BDNF高表达组3年总生存率低于BDNF低表达组(P均<0.05)。结论结肠癌组织中miR-129-5p低表达,BDNF高表达;二者表达变化与结肠癌发生发展及预后有关。 Objective To investigate the expression changes and clinical significance of microRNA-129-5p(miR-129-5p)and brain-derived neurotrophic factor(BDNF)in the colon cancer tissues.Methods A total of 118 patients with colon cancer were selected and received radical or palliative surgery.We took the colon cancer tissues and adjacent tissues(≥5 cm from the edge of the tumor)removed during the operation.The qRT-PCR method was used to detect the expression of miR-129-5p and BDNF mRNA in the tissues;Spearman correlation method was used to analyze the correlation between them,and we analyzed the relationship between the expression changes of the two and the clinicopathological characteristics of colon cancer patients.Patients were followed up,and the 3-year overall survival rate was recorded.Meanwhile,we analyzed the relationship between miR-129-5p,BDNF mRNA expression changes and patients’prognosis.Results Compared with the adjacent tissues,the relative expression of miR-129-5p in the colon cancer tissues was lower,and the relative expression of BDNF was higher(both P<0.05).The expression levels of miR-129-5p and BDNF mRNA were related to the degree of differentiation of colon cancer, TNM staging, depth of invasion, and lymph node metastasis (all P<0.05), but not related to the patient's gender, age, or tumor diameter (all P>0.05). There was a negative correlation between miR-129-5p and BDNF mRNA expression in the colon cancer tissues (rs=-5.174, P<0.05). Based on the median expression of miR-129-5p and BDNF mRNA in colon cancer tissues, colon cancer patients were divided into the high and low expression groups;the 3-year overall survival rate of the high miR-129-5p expression group was higher than that of the low miR-129-5p expression group, and the 3-year overall survival rate of the high BDNF expression group was lower than that of the low BDNF expression group (both P<0.05). Conclusion The miR-129-5p is low expressed and BDNF is highly expressed in the colon cancer tissues;the expression changes of both of them are related to the development and progression of colon cancer patients.
