丙泊酚对胃癌细胞恶性生物学行为的影响及机制 被引量:3

摘要 目的探讨丙泊酚对胃癌细胞增殖、侵袭、迁移的影响及机制。方法将培养至对数生长期的人胃癌细胞株MGC-803随机分为0、2、4、8μg/L丙泊酚组并用相应浓度丙泊酚进行干预,其中0μg/L丙泊酚组为对照组。用MTT法检测细胞增殖能力,用Transwell小室实验检测细胞侵袭能力,用细胞划痕实验检测细胞迁移能力,用Western blotting法检测细胞外信号调节激酶(ERK)、血管内皮生长因子(VEGF)蛋白表达。结果与对照组比较,各丙泊酚组细胞存活率低、侵袭至小室下室的细胞数目少、细胞划痕愈合率低(P均<0.05),且以上变化呈浓度依赖性(P均<0.05)。结论丙泊酚可抑制胃癌细胞增殖、侵袭及迁移,这可能与ERK/VEGF信号通路有关。 Objective To investigate the effects and mechanism of propofol on proliferation,invasion,and migration of gastric cancer cells.Methods The human gastric cancer cell line MGC-803 cultured to logarithmic phase was randomly divided into 0,2,4 and 8μg/L propofol groups which were separately intervened by the corresponding concentrations of propofol,and the 0μg/L propofol group was taken as the control group.MTT assay was used to detect cell proliferation,Transwell assay was used to detect cell invasion,Scratch assay was used to detect cell migration,and Western blotting was used to detect the expression of extracellular signal-regulated kinase(ERK)and vascular endothelial growth factor(VEGF).Results Compared with the control group,the cell survival rate,the number of cells invading the subventricular chamber and the wound healing rate were lower in each propofol group(all P<0.05),and the above changes were concentration-dependent(all P<0.05).Conclusion Propofol can inhibit the proliferation,invasion and migration of gastric cancer cells,which may be related to ERK/VEGF signaling pathway.
