摘要
目的探讨胰腺癌组织中miR-338-3p、β-1,3-N-乙酰氨基葡萄糖氨基转移酶3(B3GNT3)的表达变化及意义。方法选择行腹腔镜切除手术的胰腺癌患者92例,术中取癌组织及其癌旁组织(距肿瘤组织边缘≥5 cm),采用qRT-PCR检测组织中miR-338-3p、B3GNT3 mRNA表达,用Pearson相关分析二者相关性,分析二者与患者临床病理特征的关系;对患者进行随访,统计患者3年生存率,分析miR-338-3p、B3GNT3 mRNA表达与胰腺癌患者生存的关系;Cox回归分析影响胰腺癌患者生存的危险因素。结果与癌旁组织比较,胰腺癌组织中miR-338-3p表达低,B3GNT3 mRNA表达高(P均<0.05)。胰腺癌组织中miR-338-3p表达与B3GNT3 mRNA表达呈负相关(r=-0.571,P<0.05)。新发糖尿病、低中分化、TNM分期Ⅲ期、有淋巴结转移的胰腺癌患者miR-338-3p低于无新发糖尿病、高分化、TNM分期Ⅰ~Ⅱ期、未发生淋巴结转移患者(P均<0.05),低中分化、TNM分期Ⅲ期、有淋巴结转移的胰腺癌患者B3GNT3 mRNA表达高于高分化、TNM分期Ⅰ~Ⅱ期、未发生淋巴结转移患者(P均<0.05)。根据miR-338-3p、B3GNT3 mRNA相对表达量的均值将患者分为miR-338-3p高表达组、miR-338-3p低表达组,B3GNT3高表达组、B3GNT3低表达组。miR-338-3p低表达组3年生存率低于miR-338-3p高表达组,B3GNT3低表达组3年生存率高于B3GNT3高表达组(P均<0.05)。有淋巴结转移、低表达miR-338-3p、高表达B3GNT3是胰腺癌患者术后3年死亡的危险因素(P均<0.05)。结论胰腺癌组织中miR-338-3p低表达、B3GNT3 mRNA高表达,这与胰腺癌恶性生物学行为及预后不良有关。
Objective To investigate the expression changes of miR-338-3p andβ-1,3-N-acetylglucosaminotransferase 3(B3GNT3)in the pancreatic cancer tissues and their significance.Methods Ninety-two cases of pancreatic cancer patients underwent laparoscopic resection were selected.The tumor tissues and their adjacent tissues(≥5 cm from the edge of tumor tissues)were collected intraoperatively,and the expression levels of miR-338-3p and B3GNT3 mRNA in the tissues were detected by real-time quantitative PCR.Pearson correlation was used to analyze the correlation between the miR-338-3p and B3GNT3 mRNA and the clinicopathological characteristics of patients.The 3-year survival rate of the patients was statistically analyzed,and the relationship between miR-338-3p and B3GNT3 mRNA expression and the survival of pancreatic cancer patients was analyzed.Cox regression was used to analyze the risk factors for survival in pancreatic cancer patients.Results Compared with the adjacent tissues,the expression of miR-338-3p was lower and the expression of B3GNT3 mRNA was higher in the pancreatic cancer tissues(all P<0.05).The expression of miR-338-3p was negatively correlated with the expression of B3GNT3 mRNA in pancreatic cancer tissues(r=-0.571,P<0.05).MiR-338-3p in pancreatic cancer patients with emerging diabetes,low and moderate differentiation,TNM stageⅢstage,and lymph node metastasis was lower than that of patients with no emerging diabetes,high differentiation,TNMⅠ-Ⅱstage,and no lymph node metastasis(all P<0.05).The expression of B3GNT3 mRNA in patients with low and moderate differentiation,TNM stageⅢand lymph node metastasis was higher than that in patients with high differentiation,TNMⅠ-Ⅱstage and no lymph node metastasis(all P<0.05).According to the mean relative expression levels of miR-338-3p and B3GNT3 mRNA,the patients were divided into the high miR-338-3p expression group,the low miR-338-3p expression group,the high B3GNT3 mRNA expression group and the low B3GNT3 mRNA expression group.The 3-year survival rate of the group with low miR-338-3p expression was lower than that of the group with high miR-338-3p expression,and that of the group with low B3GNT3 mRNA expression was higher than that of the group with high B3GNT3 mRNA expression(all P<0.05).Lymph node metastasis,low expression of miR-338-3p and high expression of B3GNT3 mRNA were risk factors for death 3 years after surgery in pancreatic cancer patients(all P<0.05).Conclusion The low expression of miR-338-3p and high expression of B3GNT3 mRNA in pancreatic cancer tissues are related to the malignant biological behavior and poor prognosis of pancreatic cancer.
