谷氨酸草酰乙酸转氨酶抑制脑损伤后炎症反应的研究

摘要 目的研究颅脑损伤(TBI)大鼠模型脑组织中炎性细胞因子(TNF-α、IL-6)的时间依从性变化,并探讨谷氨酸草酰乙酸转氨酶(GOT)干预后的影响。方法随机选取健康成年雄性SD大鼠,应用随机数字表法分为假手术组(Sham组)、颅脑损伤组(TBI组)及TBI+谷氨酸草酰乙酸转氨酶处理组(TBI+GOT组),采用改良FeeneyDM自由落体撞击法制备TBI模型,Sham组开颅窗后不予以撞击;TBI+GOT组于制模前经颈部皮下注射GOT1周;分别于模型制作成功后12h、1d、3d、5d处死大鼠,制备脑组织匀浆,采用酶联免疫吸附试验(ELISA)检测肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)的表达情况。结果与Sham组比较,TBI组和TBI+GOT组脑组织TNF-α和IL-6的浓度在脑损伤后12h、24h显著升高(P<0.05),第3、5天无显著变化;与TBI组比较,TBI+GOT组脑组织TNF-α和IL-6的浓度在脑损伤后12h、24h显著降低(P<0.05),第3、5天无显著变化。结论颅脑损伤后脑组织炎性细胞因子(TNF-α、IL-6)的释放具有时间依从性,谷氨酸草酰乙酸转氨酶处理后炎性因子的释放受到抑制。 Objective To study the time-dependent changes of inflammatory factors(TNF-α,IL-6)in the brain tissue of trau⁃matic brain injury(TBI)rat models,and to explore the effect of Glutamate oxaloacetate transaminase(GOT)intervention.Methods Healthy adult male SD rats were randomly selected and divided into sham operation group(Sham group),traumatic brain injury group(TBI group)and TBI+glutamate oxaloacetate transaminase treatment group(TBI+GOT group)using random number table meth⁃ods),the TBI model was prepared by the modified Feeney DM free-fall impact method,the Sham group was not impacted after the craniotomy window;in the TBI+GOT group,GOT was injected subcutaneously into the neck for 1 week before model making;rats were sacrificed at 12h,1d,3d,and 5d after the model was successfully made,enzyme-linked immunosorbent assay(ELISA)was used to de⁃tect the expression of inflammatory factors tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6).Results Compared with the Sham group,the concentration of IL-6 and TNF-αin the brain tissue of the TBI group and TBI+GOT group was significantly in⁃creased at 12 hours and 24 hours after the brain injury(P<0.05),and there was no significant change on the third and fifth days;com⁃pared with the TBI group,the concentration of IL-6 and TNF-αin the brain tissue of the TBI+GOT group decreased significantly at 12 hours and 24 hours after the brain injury(P<0.05),and there was no significant change on the third and fifth day.Conclusion After traumatic brain injury,the release of inflammatory cytokines(TNF-α,IL-6)in brain tissue is time-dependent,and the release of inflammatory factors is inhibited after GOT treatment.
