RNA结合蛋白相关肝细胞癌预后模型的建立与验证

摘要 目的探究RNA结合蛋白(RBP)在肝细胞癌(HCC)中的表达及对患者预后的影响。方法从癌症基因组图谱数据库(TCGA)下载HCC的RNA序列数据,测定正常组织和肿瘤组织中差异表达的RBP水平,通过单因素、多因素回归分析筛选影响HCC患者预后的独立RBP,并构建预后模型。结果基于EIF2AK4、PRKDC、DHX37、INTS8和EEF1E1建立的预后模型,与低风险组相比,高风险组患者的总生存率低,差异有统计学意义(P<0.05)。模型受试者工作曲线的曲线下面积在建模组中为0.710,在验证组中为0.660。结论基于EIF2AK4、PRKDC、DHX37、INTS8和EEF1E1 RBP建立的预后模型,不仅能评估HCC患者预后,还能助于临床鉴别诊断。 Objective To investigate the expression of RNA-binding protein(RBP)in hepatocellular carcinoma(HCC)and its effect on the prognosis of patients.Methods The RNA sequence data of HCC were downloaded from the Cancer Genome Atlas(TCGA).The differentially expressed RBP levels in normal and tumor tissues were measured.The independent RBP saffecting the prognosis of HCC patients were screened by univariate and multivariate regression analysis,and the prognosis model was constructed.Results The prognostic model based on EIF2AK4,PRKDC,DHX37,INTS8 and EEF1E1 showed that the overall survival rate of patients in the high-risk group was lower than that in the low-risk group(P<0.05).The area under the curve of the working curve of model subjects was 0.710 in the modeling group and 0.660 in the validation group.Conclusion The prognostic model based on EIF2AK4,PRKDC,DHX37,INTS8 and EEF1E1 RBP can not only evaluate the prognosis of HCC patients,but also contribute to clinical differential diagnosis.
