摘要
器官移植是治疗人脏器功能衰竭的理想选择之一,但是器官来源的短缺明显影响该技术在临床的应用。目前即使运用扩大供体来源的多种措施以增加器官供体来源,也不能从根本上解决供受体不平衡的状况。扩大供器官来源成为移植工作者永恒的话题,而解决器官供体来源不足最好的方法是异种移植。但因物种进化之间的差异,异种移植暂未能进入临床,因此目前一直进行猪--非人灵长类动物的临床前研究。猪因解剖及生理与人相近及容易进行基因编辑等优点,成为良好潜在的供体来源。随着基因编辑技术越来越娴熟,多基因修饰器官猪成为可能,为临床选择更优基因组合供体提供了技术保障。本文主要阐述人膜辅助蛋白在异种移植中的作用。
Organ transplantation is one of the ideal options for the treatment of human organ failure,but the shortage of organ sources obviously affects the clinical application of this technique.At present,even if various mea-sures are adopted to expand the donor source and increase the organ donor source,the imbalance between donor and re-cipient cannot be solved fundamentally.Expansion of donor organ sources has become an eternal topic for transplant workers,and xenotransplantation is the best way to solve the shortage of donor organ sources.Due to differences in the evolution of species,xenotransplantation has not been introduced into clinical practice for the time being,so preclinical studies on porcine and non-human primates have been carried out.Pigs have become a good potential donor source due to their anatomical and physiological similarities to humans and the ease of gene editing.With the development of gene editing technology more and more proficient,it becomes possible for pigs with multiple genetically modified organs,which provides a technical guarantee for the clinical selection of better gene combination donors.This article mainly dis-cusses the role of human membrane cofactor proteins in xenotransplantation.
