Ni离子螯合亲和层析法分离大叶海藻中的抗氧化多肽

Separation of Antioxidant Polypeptides from Seaweed by Ni Chelating Affinity Chromatography

摘要 采用Ni2+螯合抗氧化多肽,经亲和层析柱分离纯化的方法提取大叶海藻中的抗氧化多肽。对影响洗脱效果和抗氧化多肽乳化活性的因素进行探讨,并分析抗氧化多肽的洗脱曲线、组成成分和螯合率。对提取的抗氧化多肽的活性在O_(2)-·和·OH清除能力2个方面进行测定。结果表明,Ni离子螯合多肽的亲和层析分离方法有效,可获得较好的多肽纯度和活性。 Antioxidant polypeptide was extracted from marine algaeby Ni2+chelated antioxidant polypeptide and separated and purified by affinity chromatography column.The factors influencing elution efficiency and emulsifying activity of antioxidant polypeptide were discussed.Elution curve,composition and chelating rate of antioxidant polypeptide were analyzed.The activity of the antioxidant polypeptide was determined in two aspects:the scavenging ability of O_(2)-·and·OH.The results showed that the affinity chromatography method of Ni2+chelated polypeptide was effective,and it could achieve good purity and activity performance of the polypeptide.
