Extraction of Polysaccharide from Ganoderma Lucidum and Its Antioxidant Activity by Steam Explosion Combined with Ultrasonic

摘要 以灵芝子实体原料,在单因素试验基础上,采用正交试验优化确定蒸汽爆破对灵芝子实体粗多糖预处理的最佳工艺,以DPPH自由基清除率和还原力为指标,研究蒸汽爆破处理对灵芝子实体粗多糖抗氧化活性的影响。结果表明,蒸汽爆破最佳工艺为稳压时间100 s、蒸汽爆破压强1.3 MPa、超声时间20 min。在此条件下,灵芝子实体粗多糖得率为14.21%,是未处理的样品多糖得率1.86倍。扫描电镜分析显示蒸汽爆破使灵芝子实体表面纤维束断裂,出现空隙,可能导致多糖提取率提高。体外抗氧化试验表明,经过蒸汽爆破处理后DPPH和羟自由基清除率IC50分别提高16%和31%,总还原力较未处理子实体样品低略低。试验分析蒸汽爆破技术应用于灵芝子实体多糖提取的可行性,并寻求一种有效的提取方法。 On the base of single-factor experiment,taking ganoderma lucidum as material,the extraction technological parameters for crude polysaccharide optimized by orthogonal test with the pretreatment of steam explosion were measured.Taking DPPH radical scavenging rate and reduction force as indices,the effect of steam explosion treatment on antioxidant activity of crude polysaccharide from Ganoderma lucidum fruit body was studied.The optimum technological parameters were steam explosion pressure-holding time 100 s,pressure 1.3 MPa,and ultrasonic time 20 min,with the crude polysaccharide receiving rate 14.21%,which was 1.86 times that of untreated samples.Scanning electron microscope(SEM)analysis showed that the steam explosion broke the fiber bundles on the solid surface of Ganoderma lucidum and voids appeared,which might lead to the increase of the extraction rate of polysaccharides.In vitro anti-oxidation test,DPPH and Hydroxyl radical scavenging rate IC50 increased by 16%and 31%after steam blasting treatment,but the reduction force was lower than that of untreated samples.The feasibility of steam blasting technology in extracting polysaccharide from Ganoderma lucidum was analyzed,to find an more effective extraction method.
