摘要
建立阿胶及其保健食品中马源性成分环介导等温扩增检测方法,对市售阿胶及其保健食品进行马源性成分检测分析。特异性引物选用马的线粒体ATPase 6基因,优化可视化LAMP反应条件,验证引物的特异性和灵敏度。可视化LAMP对阿胶及其保健食品中马源性成分的灵敏度为0.0001 ng/μL,是实时荧光PCR法的100倍。该检测方法操作简便、专属性强、灵敏度高,适合于阿胶及其保健食品生产企业和基层市场监督管理部门。
A method of loop-mediated isothermal amplification for detection of horse derived components in Asini corii Colla and health foods was developed.Specific primers were designed for the mitochondrial ATPase 6 gene of the horse,and the LAMP reaction conditions were optimized to verify the specificity of the primers.The sensitivity of visual LAMP to equine compounds in Asini corii Colla and health foods was 0.0001 ng/μL,which was 100 times that of real-time PCR.The method was simple,specific and sensitive,and was suitable for Asini Corii Colla and health foods enterprises and grass roots market supervision and management departments.
