甘草胚性愈伤组织的诱导与增殖

摘要 [目的]研究不同激素组合对甘草愈伤组织形成的影响,诱导甘草胚性愈伤组织的分化与增殖。[方法]以甘草种子为试验材料,通过种子发芽率比较,筛选出发芽率最高的浸种条件。利用植物组织与细胞培养技术,进行愈伤组织诱导与增殖研究,通过调节激素组合对甘草愈伤组织形成的影响进行研究。[结果]浸种40 min条件下,种子发芽率最高达64%;下胚轴为最适合诱导愈伤组织的外植体;MS+6-BA 1.0 mg/L+NAA 2.0 mg/L为甘草愈伤组织增殖培养最适培养基;MS+NAA 2.0 mg/L+6-BA 0.5 mg/L有利于甘草生根培养物胚性愈伤组织的形成;MS+NAA0.3 mg/L+6-BA 1.5 mg/L有利于甘草生芽培养物胚性愈伤组织的形成;MS+NAA 0.1 mg/L+6-BA 1.0 mg/L时胚性愈伤组织增殖培养为最适,增殖率为378.0%。[结论]甘草外植体诱导愈伤组织的适宜程度为下胚轴>胚根>带芽茎段>子叶。相比NAA与KT激素组合,NAA与6-BA激素组合更适宜甘草胚性愈伤组织增殖,且达到最大增殖率为378.0%。 [Objective]To study the effects of different hormone combinations on callus formation of Glycyrrhiza uralensis,and to induce differentiation and proliferation of embryogenic callus of Glycyrrhiza uralensis.[Method]Taking Glycyrrhiza uralensis seeds as experimental materials,the soaking conditions with the highest germination rate were screened out by comparing the germination rates of seeds.Using plant tissue and cell culture technology,callus induction and proliferation were studied,and the effect of adjusting hormone combination on callus formation of Glycyrrhiza uralensis was studied.[Result]The germination rate of seeds reached 64%under the condition of soaking seeds for 40 minutes;hypocotyl is the most suitable explant for callus induction;MS+6-BA 1.0 mg/L+NAA 2.0 mg/L is the most suitable medium for proliferation culture of Glycyrrhiza uralensis callus.The MS+NAA 2.0 mg/L+6-BA 0.5 mg/L was beneficial to the formation of embryogenic callus of Glycyrrhiza uralensis rooting culture.MS+NAA 0.3 mg/L+6-BA 1.5 mg/L was beneficial to the formation of embryogenic callus of Glycyrrhiza uralensis bud culture.Embryogenic callus was the best when MS+NAA 0.1 mg/L+6-BA 1.0 mg/L,and the proliferation rate was 378.0%.[Conclusion]The suitable degree of callus induction from Glycyrrhiza uralensis explants is hypocotyl>radicle>bud stem>cotyledon.Compared with the combination of NAA and KT hormone,the combination of NAA and 6-BA hormone is more suitable for the proliferation of embryogenic callus of Glycyrrhiza uralensis,and the maximum proliferation rate is 378.0%.
