摘要
目的研究3-{[(叔丁基二甲基硅基)氧基]甲基}环丁烷-1-羧酸的合成工艺路线。方法以3-亚甲基环丁基腈为起始原料,经过水解、还原、硅醚保护、硼氢化氧化、氧化等实验操作合成3-{[(叔丁基二甲基硅基)氧基]甲基}环丁烷-1-羧酸。结果合成了目标化合物,并利用NMR、HR-MS确证了结构。结论3-{[(叔丁基二甲基硅基)氧基]甲基}环丁烷-1-羧酸合成工艺操作性好,条件温和,无需贵重试剂和柱色谱操作,适于工业化生产。
Objective To study the synthetic routes of 3-{[(tert-butyldimethylsilyl)oxy]methyl}cyclobutane-1-carboxylic acid.Methods With 3-methylenecyclobutane-1-carbonitrile as the starting material,3-{[(tert-butyldimethylsilyl)oxy]methyl}cyclobutane-1-carboxylic acid was synthesized through optimized synthetic procedures by using tipical synthetic operations including:hydrolysis,reduction,protection,oxidation.Results The target molecule was successfully synthesized and the structure was confirmed by NMR and HR-MS spectroscopy.Conclusion The synthesis process of 3-{[(tert-butyldimethylsilyl)oxy]methyl}cyclobutane-1-carboxylic acid has good operability,mild conditions without using expensive transion metals and column-purification,which is quite suitable for industrial production.
