摘要
目的探讨硫氧还蛋白(thioredoxin,Trx)与硫氧还蛋白相互作用蛋白(thioredoxin-interacting protein,TXNIP)在结肠息肉、结肠腺瘤及结肠癌中的表达,分析Trx、TXNIP在结肠息肉癌变中的作用。方法收集河北北方学院附属第一医院2019年1月~2019年10月就诊的结肠息肉(结肠息肉组)20例、结肠腺瘤(结肠腺瘤组)40例、结肠癌(结肠癌组)20例以及正常结肠(正常结肠组)20例,通过ELISA法检测血清Trx、TXNIP的含量,采用胰岛素还原法检测血清Trx活性,苏木精-伊红染色(HE)法观察各组织的形态学,应用免疫组织化学法检测组织中Trx、TXNIP的表达,实时荧光定量PCR检测组织中Trx mRNA与TXNIP mRNA的表达水平。结果与正常结肠组比较,结肠腺瘤组与结肠癌组患者血清中Trx含量和活性明显升高,结肠息肉组、结肠腺瘤组患者与结肠癌组患者血清中TXNIP含量较正常结肠组明显下降,差异均有统计学意义(P<0.05);在结肠腺瘤组和结肠癌组组织中Trx阳性表达率明显高于正常结肠组,而TXNIP阳性表达率明显低于正常结肠组,差异均有统计学意义(P<0.05);与正常结肠组比较,结肠息肉组、结肠腺瘤组及结肠癌组组织中Trx mRNA的表达水平均明显上调,TXNIP mRNA的表达水平均明显下调,差异具有统计学意义(P<0.05)。结论Trx在结肠息肉癌变中高表达,而TXNIP表达降低,表明两者都参与结肠癌前病变与癌组织的发生及发展,两者均在结肠息肉癌变过程中起着重要的作用。
Objective To study the expression of thioredoxin(Trx)and thioredoxin interacting protein(TXNIP)in colon polyps,colon adenoma and colon cancer and explore the role of Trx and TXNIP in the carcinogenesis of colon polyps.Methods 20 cases of colonic polyps(colonic polyps group),40 cases of colonic adenomas(colonic adenomas group),20 cases of colon cancer(colon cancer group)and 20 cases of normal colon(normal colon group)were collected from the First Affiliated Hospital of Hebei North University from January 2019 to October 2019.The contents of serum TRX and TXNIP were detected by ELISA,and the activity of serum TRX was detected by insulin reduction method,The morphology of each tissue was observed by hematoxylin eosin staining(HE).The expressions of TRX and TXNIP were detected by immunohistochemistry.The expression levels of TRX mRNA and TXNIP mRNA were detected by real-time fluorescence quantitative PCR.Results Compared with the normal colon group,the Trx content and activity in the serum of colon adenoma patients and colon cancer patients were significantly increased,the TXNIP content in the serum of colon polyps,colon adenoma and colon cancer patients was significantly lower than that in the normal colon group,the difference is statistically significant.Academic significance(P<0.05).The positive expression rate of Trx in colon adenoma tissue and colon cancer tissue was significantly higher than that in normal colon tissue,while the positive expression rate of TXNIP was significantly lower than normal colon tissue,the difference was statistically significant(P<0.05).Compared with normal colon tissue,the expression level of Trx mRNA in colon polyp tissue,colon adenoma tissue and colon cancer tissue was significantly up-regulated,and the expression level of TXNIP mRNA was significantly down-regulated,and the difference was statistically significant(P<0.05).Conclusion The expression of Trx and TXNIP in the carcinogenesis of colon polyps gradually increases,and they participate in the occurrence and development of colon precancerous lesions and cancer tissues.Both of them play an important role in the carcinogenesis of colon polyps.
