叶用莴苣叶绿体型谷氨酰胺合成酶基因的克隆及不同形态氮素配比下的表达分析

摘要 为探讨不同形态氮素配比(NO3-:NH4+)对叶用莴苣CGS基因表达的影响,以叶用莴苣为材料,克隆了叶用莴苣CGS基因cDNA编码序列(CDS)全长,并对其进行了生物信息学分析和表达分析。结果表明:(1)叶用莴苣CGS基因cDNA的CDS全长为1293bp,编码氨基酸数量为430;其编码蛋白分子式为C_(2095)H_(3252)N_(572)O_(637)S_(14),属亲水性蛋白和无信号肽;(2)随着铵态N比例的增加,叶用莴苣CGS基因的表达亦呈先升后降趋势,铵态N比例为25%时最高,各处理皆在采样第4天达到最大值。说明在不同形态氮素配比下,叶用莴苣CGS的表达在氮同化反应中的起主导作用。
