产纤维素酶细菌的筛选及其产酶优化 被引量:3

摘要 利用刚果红染色法筛选产纤维素酶菌株,采用分子生物学技术和形态学观察作为鉴定手段,通过滤纸条崩解实验测定实际降解能力。以葡聚糖内切酶活(CMCase)和滤纸酶活(FPase)为指标,进行单因素和响应面试验优化。结果表明,获得一株产纤维素酶细菌(Xh-12),鉴定为不动杆菌属,发酵培养7 d后对滤纸条的减重率可达54.4%。在接种量1.5%、温度39℃、pH=6.7的最优培养条件下,CMCase活力为1.05 U/mL,FPase活力为0.76 U/mL,具有较高酶活力。 Congo red staining method was used to screen cellulase-producing strains,molecular biology technology and morphological observation were used as indexes,and the actual degradation ability was determined through the filter paper strip disintegration experiment.Using endoglucanase activity(CMCase)and filter paper enzyme activity(FPase)as indicators,single factor and response surface experiments were optimized.A cellulase-producing bacterium(Xh-12)was obtained from this experiment,and the results showed that it was acinetobacter.The weight loss rate of the filter paper strips can reach 54.4%after 7 days of fermentation culture.Under the optimal culture conditions of 1.5%inoculum,temperature 39℃,and pH=6.7,CMCase activity was 1.05 U/mL and FPase activity was 0.76 U/mL,which showed high enzyme activity.
