基于聚类分析和主成分分析的广西小叶金花草高效液相色谱指纹图谱研究 被引量:10

摘要 目的建立小叶金花草的高效液相色谱(HPLC)指纹图谱,综合评价其质量。方法采用“中药色谱指纹图谱相似度评价系统”建立广西产小叶金花草的HPLC指纹图谱,色谱柱为Agilent XBD-C_(18),流动相为乙腈-0.4%磷酸溶液(梯度洗脱),流速为1.0 mL·min^(-1),检测波长为260 nm,柱温为25℃,用对照品定性部分共有峰,并采用SPSS软件进行聚类分析和主成分分析。结果共确定了11个共有峰,指认了其中4个共有成分(1、4、5、8号共有峰分别为原儿茶酸、香草酸、咖啡酸和菊苣酸);13批药材可聚为三类;提取出3个主成分,累积方差贡献率达到84.643%。结论该方法简便、快捷,结合聚类分析和主成分分析可为药材的质量评价提供参考。 Objective To establish an HPLC fingerprint of Onychium japonicum(Thunb.)Kze.to evaluate its quality comprehensively.Methods HPLC fingerprints of Onychium japonicum from Guangxi were established by Similarity Evaluation System Chromatographic Fingerprint of TCM software,and detected by Agilent XDB-C_(18)column with mobile phase consisting of acetonitrile-0.4%phosphoric acid solution(gradient elution)at the flow rate of 1.0 mL·min^(-1).The detection wavelength was 260 nm,and the column temperature was 25℃.Some common peaks were identified by reference substances.Cluster analysis and principal component analysis were performed by SPSS software.Results Totally 11 common peaks were determined,4 common peaks(peak 1,4,5,8)were indentified as protocatechuic acid,vanillic acid,caffeic acid and cichoric acid;13 batches of medicinal materials clutered into 3 groups;3 principal components were extracted and the cumulative contribution rate was 84.643%.Conclusion This method is simple and rapid,which can provide reference for the quality evaluation of Onychium japonicum(Thunb.)Kze.from Guangxi combining the cluster analysis and principal component analysis.
