摘要
目的比较紫外分光光度法测定血管瘤乳膏中马来酸噻吗洛尔含量的4种定量分析方法。方法采用标准曲线法(298 nm)、吸收系数法(298 nm)、双波长法(测定波长298 nm、参比波长330 nm)、一阶导数法(316 nm)对紫外扫描光谱数据进行定量分析,并对4种方法进行方法学考察。结果标准曲线法(298 nm)、吸收系数法(298 nm)、双波长法(测定波长298 nm、参比波长330 nm)、一阶导数法(316 nm)中马来酸噻吗洛尔含量线性范围为10~40μg·mL^(-1)(r≥0.9994),平均回收率为94.6%~97.1%,日内精密度RSD分别为1.6%、1.6%、1.6%、0.0%;日间精密度RSD分别为2.2%、2.6%、1.3%、0.0%。结论4种分析方法都可用于血管瘤乳膏中马来酸噻吗洛尔的含量定量分析,其中双波长法和一阶导数法更为简便。
Objective To compare 4 quantitative methods in the determination of timolol maleate in hemangioma cream with UV spectrophotometer.Methods UV scanning spectrum was analyzed quantitatively by standard curve method(298 nm),absorption coefficient(298 nm),dual-wavelength spectrophotometry(detection wavelength of 298 nm,reference wavelength of 330 nm)and firstderivative spectrophotometry(316 nm).Methodology of 4 methods were compared.Results The linearity of timolol maleate was 10~40μg·mL^(-1)(r≥0.9994)by standard curve method(298 nm),absorption coefficient(298 nm),dual-wavelength spectrophotometry(detection wavelength of 298 nm,reference wavelength of 330 nm)and first-derivative spectrophotometry(316 nm)with average recoveries of 94.6%~97.1%,RSDs were 1.6%,1.6%,1.6%,and 0.0%for intra-day precision,and 2.2%,2.6%,1.3%,and 0.0%for inter-day precision.Conclusion All the 4 methods can be used for the determination of timolol maleate in hemangioma cream.The dual-wavelength spectrophotometry and first-derivative spectrophotometry are simpler and more convenient.
