生物信息学预测丹参酮ⅡA影响乳腺癌预后的相关基因

摘要 目的通过生物信息学方法分析与丹参酮ⅡA影响乳腺癌发展相关的差异基因。方法检索基因表达数据库(GEO)获得GSE85871目标数据集,筛选丹参酮ⅡA作用前后乳腺癌的差异表达基因。对其进行基因本体论(GO)及京都基因和基因组百科全书(KEGG)分析,利用蛋白互作网络分析(PPI)获得核心基因后依次进行生存分析,根据P值得到具有预后价值的基因。结果GO分析示差异基因在脂多糖反应、受体聚集、终结、突触后密集区、糖蛋白结合等中富集。KEGG分析在自然杀伤细胞介导细胞毒性和Ras相关蛋白信号通路中显著富集。筛选得到神经营养受体酪氨酸激酶2(NTRK2)和CXC趋化因子配体11(CXCL11)目的基因。结论NTRK2和CXCL11基因表达与乳腺癌预后有关(P<0.05),有望为进一步探索丹参酮ⅡA作用于乳腺癌的分子机制指明方向。 Objective To analyze the differential genes related to the effects of Tanshinone IIA on progression of breast cancer via bioinformatic approach.Methods The targeted dataset GSE85871 was obtained by searching of the Gene Expression Omnibus(GEO)database,with which,the differentially expressed genes in breast cancer before and after Tanshinone IIA treatment were screened.The findings were subjected to Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analyses.The core genes involved were determined using the protein-protein interaction(PPI)network analysis,and were fed into survival analyses in successive steps.Finally,genes with prognostic value were identified according to P values.Results GO analysis showed that differentially expressed genes were enriched in response to lipopolysaccharide,receptor clustering,terminal bouton,postsynaptic density,and glycoprotein binding;by KEGG analysis,these genes were significantly enriched in natural killer cell-mediated cytotoxicity and Rap 1 signaling pathway.Finally,the target genes were screened and identified to be the neurotrophic receptor tyrosine kinase 2(NTRK2)and the C-X-C motif chemokine ligand 11(CXCL11).Conclusion The gene expression of NTRK2 and CXCL11 is related to prognosis in breast cancer(P<0.05).This may offer direction to further study on the molecular mechanism by which Tanshinone IIA acts on breast cancer.
