Expression and biological function of nuclear division cycle 80 in gliomas

摘要 目的探讨核分裂周期蛋白80(NDC80)基因在胶质瘤中的表达及其生物学功能。方法通过提取癌症基因组图谱数据库(TCGA)中胶质瘤患者的相关临床资料和测序数据,分析NDC80基因表达与胶质瘤患者临床参数及预后的关系。分析与NDC80共表达的基因,并对这些基因进行富集分析。通过敲减NDC80基因,探索NDC80表达减少对胶质瘤细胞增殖和细胞周期的影响。结果 NDC80在胶质瘤组织中的表达较正常脑组织高(1.996±1.314、0.099±0.148,P<0.01),且NDC80的高表达与不良预后相关[风险比(HR)=5.12,(P<0.01)]。单因素、多因素Cox回归分析显示NDC80是胶质瘤的独立预后因子(均P<0.01)。NDC80的共表达基因主要富集在细胞周期等通路上。此外,敲除胶质瘤细胞中的NDC80基因后,细胞的增殖能力受限,细胞周期阻滞在G0/G1期。结论胶质瘤组织中NDC80的上调是胶质瘤预后不良的独立预测因子。 Objective To explore the expression of nuclear division cycle 80(NDC80)in glioma and to investigate the biological processes of NDC80 in glioma cells.Methods The clinical data and sequencing data of glioma patients from The Cancer Genome Atlas(TCGA)database were collected and the relationship between NDC80 expression and clinical parameters was analyzed.The co-expressed genes with NDC80 were analyzed and enriched.NDC80 was knocked down to explore the effect of NDC80 on proliferation and cell cycle in glioma cells.Results The expression of NDC80 was significantly higher in glioma tissues than in normal brain tissues(1.996±1.314 vs.0.099±0.148,P<0.01),and high NDC80 expression was associated with poor survival[hazard ratio(HR)=5.12,P<0.01].Univariate and multivariate Cox regression analyses showed that high expression of NDC80 was an independent risk factor for overall survival(P<0.01).NDC80 co-expressed genes were mainly enriched in cell cycle pathway.In addition,NDC80 gene knockdown in glioma cells resulted in inhibition of proliferation and arrest of cells in the G0/G1 phase.Conclusion The up-regulation of NDC80 in glioma tissue is an independent predictor of poor prognosis in glioma,and NDC80 may be a potential biological target for the treatment of glioma.
