长链非编码RNA LINC01235对胃癌细胞迁移和侵袭的调控作用

Regulation of long non-coding RNA LINC01235 on migration and invasion of gastric cancer cells

目的检测长链非编码RNA LINC01235在胃癌细胞系中的表达并观察LINC01235对胃癌细胞迁移和侵袭的影响。方法采用Transwell小室法及划痕实验检测LINC01235对胃癌细胞迁移和侵袭能力的影响。应用蛋白质印迹法(Western blot)检测过表达LINC01235后上皮-间充质转化(EMT)关通路蛋白的表达变化。结果划痕实验显示,敲低LINC01235的实验组在划痕48 h后迁移距离显著短于对照组。Transwell实验显示,敲低LINC01235的实验组在24 h内穿膜细胞数量显著低于对照组(48.000±4.619比156.300±4.096,P<0.05)。蛋白质免疫印迹结果显示,过表达LINC01235的表达后,EMT通路相关蛋白N-钙黏蛋白及基质金属蛋白酶-2的表达水平随之升高。结论 LINC01235具有促进胃癌细胞迁移和侵袭的作用。

Objective To detect the expression of long non-coding RNA(lncRNA)LINC01235 in gastric cancer cell lines and observe the effect of LINC01235 on migration and invasion of gastric cancer cells.Methods Transwell chamber method and scratch test were used to detect the effect of LINC01235 on the migration and invasion of gastric cancer cells.Western blotting was used to detect the expression changes of epithelial-mesenchymal transition(EMT)-related pathway proteins after overexpression of LINC01235.Results The scratch test showed that the LINC01235 knockdown group had a shorter migration distance than the control group after 48 h of scratching.The Transwell experiment showed that the number of transmembrane cells in the LINC01235 knockdown group was significantly less than that in the control group within 24 h(48.000±4.619 vs.156.300±4.096,P<0.05).Western blotting results showed that after overexpression of LINC01235,the expression levels of N-cadherin and matrix metalloproteinase-2(MMP-2)related to the EMT pathway increased.Conclusion LINC01235 promotes the migration and invasion of gastric cancer cells.
