基于网络药理学探讨益气健脾中药复方治疗糖尿病胃轻瘫的作用机制

Effect and mechanism of Yiqi Jianpi traditional Chinese medicine compound in the treatment of diabetic gastroparesis by using network pharmacology

目的基于网络药理学探讨益气健脾中药复方治疗糖尿病胃轻瘫(DGP)的作用机制。方法运用中药系统药理学数据库与分析平台(TCMSP)和BATMAN数据库进行筛选,得出口服生物利用度≥30及类药性≥0.18的潜在活性成,用基因疾病关联(Disgenet)与Swisss数据库,得出益气健脾中药复方治疗DGP的潜在靶点。利用Cytoscape软件构建"益气健脾中药复方-活性化合物-潜在作用靶点"的网络,构建蛋白质-蛋白质相互作用(PPI)网络,用注释、可视化和集成发现数据库(DAVID)数据库对此复方的作用靶点进行基因本体(GO)生物功能分析和京都基因与基因组百科全书(KEGG)通路富集分析。结果益气健脾中药复方通过75个靶点治疗DGP,经GO分析得到1445条取生物学过程、57条细胞组分及62个分子功能;KEGG富集通路显示得到的关键通路主要有五羟色胺信号通路、环磷酸鸟苷-蛋白激酶G(cGMP-PKG)信号通路、环磷酸腺苷信号通路和钙信号通路等。结论益气健脾中药复方通过多成分-多靶点干扰多条信号通路发挥治疗DGP的效果。

Objective To explore the effect and mechanism of Yiqi Jianpi traditional Chinese medicine compound in the treatment of diabetic gastroparesis(DGP)based on network pharmacology.Methods TCMSP(http://tcmspw.com/tcmsp.php)and Batman database were used to screen the potential bioactive compounds with bioavailability≥30 and generic property≥0.18.Swisss database and Digenet database were used to find the potential targets of Yiqi Jianpi compound in the treatment of DGP.The network of"Yiqi Jianpi traditional Chinese medicine compound-active compound-potential target"was constructed by using Cytoscape software,and the protein-protein interaction(PPI)network was constructed,the biological function of go and the enrichment of Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway were analyzed by David database.Results The compound prescription of Yiqi Jianpi traditional Chinese medicine compound was obtained through 75 targets to treat DGP,1445 biological processes,57 cell component expression processes and 62 molecular function related processes by GO analysis.KEGG enrichment pathway showed that the key pathway was obtained,it mainly includes 5-HT signaling pathway,cGMP-PKG signaling pathway,cAMP signaling pathway and calcium signaling pathway.Conclusion Research Based on network pharmacology shows that,Yiqi Jianpi traditional Chinese medicine compound exert therapeutic effect on DGP by interfering multiple signal pathways with multiple components and multiple targets.
