摘要
目的研究海洛因成瘾小鼠肝中微量元素的变化。方法按照体重将雄性昆明小鼠随机分为2组:对照组和海洛因组,每组6只。海洛因组小鼠皮下注射海洛因溶液2次/天,连续13 d。在1~10 d每次给药剂量依次为10,15,20,25,30,35,40,45,50,55 mg·kg^(-1),10 d后以55 mg·kg^(-1)持续3 d;对照组小鼠皮下注射等量生理盐水。利用电感耦合等离子体质谱法(ICP-MS)测定海洛因成瘾小鼠肝内微量元素的含量。用正交偏最小二乘法-判别分析(OPLS-DA)与t检验确定组间的差异微量元素。结果 2组间有差异的微量元素为Ca、Fe和Zn。海洛因组肝中Ca、Fe和Zn的含量分别为(23.10±10.90),(83.60±30.90)和(20.30±7.97)μg·g^(-1);对照组肝中Ca、Fe和Zn的含量分别为(47.70±7.22),(130.00±6.51)和(34.10±5.98)μg·g^(-1)。海洛因成瘾小鼠肝中3种微量元素(Ca、Fe和Zn)含量明显降低,与对照组相比差异有统计学意义(P<0.01)。结论与对照组相比,海洛因成瘾小鼠肝中Ca、Fe和Zn三种微量元素水平下降,可能是海洛因诱导肝损伤的潜在机制。可为海洛因肝毒性机制的深入研究提供依据。
Objective To study the changes of trace elements in the liver of heroin addicted mice. Methods The male Kunming mice were randomly divided into two groups according to weight: control group and heroin group. Mice in the heroin group were injected subcutaneously with heroin solution twice a day for 13 consecutive days. From 1 to 10 d, the specific dosage for each administration was 10, 15, 20, 25, 30, 35, 40, 45, 50, 55 mg·kg^(-1), and then 55 mg·kg^(-1) lasts for 3 d. Inductively coupled plasma mass spectrometry(ICP-MS) was used to determine the content of trace elements in the liver of heroin-addicted mice. Orthogonal partial least squares-discriminant analysis(OPLS-DA) and t-test were used to determine the differences in trace elements between groups. Results The difference between the two groups of trace elements is Ca, Fe and Zn three trace elements. The contents of Ca, Fe and Zn in the liver of the heroin group were(23.10±10.90),(83.60±30.90) and(20.30±7.97) μg·g^(-1), respectively;the contents of Ca, Fe and Zn in the liver of the control group were(47.70±7.22),(130.00±6.51) and(34.10±5.98) μg·g^(-1), respectively. The content of three trace elements Ca, Fe and Zn in the liver of heroin-addicted mice was significantly reduced, and the difference were statistically significant compared with the control group( P < 0. 01). Conclusion Compared with the control group,the levels of the three trace elements Ca,Fe and Zn in the liver of heroin-addicted mice were decreased. It could provide a basis for in-depth research on the mechanism of heroin hepatotoxicity.
