藏药十二味翼首散治疗“年仍奈”潜在机制的网络药理学研究 被引量:2

摘要 目的通过网络药理学技术预测十二味翼首散治疗"年仍奈"的作用靶点及通路,分析其抗细胞因子风暴的多功效-多靶点-多通路的协同作用机制。方法利用文献挖掘、TCMSP和化学专业数据库预测十二味翼首散化合物及候选靶点,通过STRING 11.0数据库富集十二味翼首散靶点的GO和KEGG通路,运用GeneCards数据库预测新型冠状病毒肺炎的候选靶点,取十二味翼首散靶点与新冠肺炎靶点交集结果生成VENN图,通过STRING 11.0数据库和Cytoscape 3.6.0软件构建交集成分靶点蛋白互作网络,富集其GO功能注释和KEGG通路。采用Cytoscape 3.6.0软件构建十二味翼首散治疗"年仍奈"的"药物-活性成分-靶点-通路-功效"网络结构图。结果十二味翼首散通过作用于TNF、IL6、GAPDH、TP53、MAPK3、ALB、EGFR等核心靶点,调节免疫应答功能和炎症反应、病毒感染和血管通透及内皮细胞功能等信号通路,发挥了集热平稳、散热驱邪,抑制传播、安抚身心,调养身体的重要作用。结论本研究探讨了十二味翼首散抗细胞因子风暴的潜在机制及防治"年仍奈"的不同阶段,为十二味翼首散的临床应用提供科学依据。 Objective To predict the targets and pathways of the Shi-Er-Wei-Yi-Shou-San in the treatment of"Nian Reng Nai",and analyze the multi-component-multi-target-multi-pathway synergy mechanism of its anti-cytokine storm by network pharmacology technology.Methods Literature mining,TCMSP and Chemistry Database was used to predict the compounds and candidate targets of Shi-Er-Wei-Yi-Shou-San.The GO and KEGG pathways of the Shi-ErWei-Yi-Shou-San targets were enriched through the STRING 11.0 database,and GeneCards database was used to predict the candidate targets of new coronavirus pneumonia.A VENN map was generated by taking the intersection of Shi-Er-Wei-Yi-Shou-San targets and new coronary pneumonia targets.A cross-integrated target protein interaction network was built with STRING 11.0 database and Cytoscape 3.6.0 software,and the"Drug-Compounds-TargetPathway-Efficacy"network diagram of Shi-Er-Wei-Yi-Shou-San was constructed by Cytoscape 3.6.0 software.Results By acting on core targets such as TNF,IL6,GAPDH,TP53,MAPK3,ALB,EGFR,Shi-Er-Wei-Yi-Shou-San may regulate immune response and inflammation,viral infection,vascular permeability and endothelial cell function.It plays an important role in stabilizing heat collection,dissipating heat and dispelling evil,inhibiting spread,soothing the body and mind,and recuperating the body.Conclusion This study explored the potential mechanism of anti-cytokine storm of Shi-Er-Wei-Yi-Shou-San and the different stages of prevention and treatment of"Nian Reng Nai",provided scientific basis for the clinical application of Shi-Er-Wei-Yi-Shou-San.
