摘要
急性肠缺血再灌注损伤导致一系列的病理生理学改变,如白细胞黏附、钙超载、炎症反应、细胞凋亡、免疫反应、氧化应激等,导致全身炎症反应。间充质干细胞具有低免疫调节、免疫原性和自我更新、多能分化、抗炎特异性的特性。在免疫耐受中,B7-H1起着重要作用,是免疫治疗中有效的靶点,调节外周组织中的B细胞或T细胞应答,反向调节免疫应答及自身免疫疾病。间充质干细胞上调B7-H1的表达,增加抑炎因子的表达,增加免疫调节作用,抑制促炎因子的表达。急性肠缺血再灌注损伤后,间充质干细胞可增强肠组织的抗氧化能力,抗炎细胞因子,减少炎性细胞因子,B7-H1信号通路参与间充质干细胞对肠缺血再灌注损伤的保护。
Acute intestinal ischemia-reperfusion injury leads to a series of pathophysiological changes,such as leukocyte adhesion,calcium overload,inflammatory response,cell apoptosis,immune response,oxidative stress,etc.,leading to systemic inflammation.Mesenchymal stem cells have the characteristics of low immune regulation,immunogenicity and self-renewal,pluripotent differentiation,and anti-inflammatory specificity.In immune tolerance,B7-H1 plays an important role and is an effective target in immunotherapy.It regulates B cell or T cell responses in peripheral tissues and reversely regulates immune responses and autoimmune diseases.Mesenchymal stem cells up-regulate the expression of B7-H1,increase the expression of anti-inflammatory factors,increase immune regulation,and inhibit the expression of pro-inflammatory factors.After acute intestinal ischemia-reperfusion injury,mesenchymal stem cells can enhance the antioxidant capacity of intestinal tissue,anti-inflammatory cytokines,and reduce inflammatory cytokines.The B7-H1 signaling pathway is involved in the protection of mesenchymal stem cells against intestinal ischemia-reperfusion injury.
