基于网络药理学探讨石榴健胃丸治疗胃肠炎的作用机制 被引量:2

摘要 目的利用网络药理学探讨石榴健胃丸治疗胃肠炎的作用靶点及信号通路,阐述其可能的作用机制。方法利用TCMSP和TCMID数据库,以OB≥30%,DL≥0.18为条件筛选石榴健胃丸中的活性成分和作用靶点。基于GeneCards和Dis-GeNet数据库获取胃肠炎相关靶点,蛋白质相互作用网络通过String数据库构建。成分-靶点-疾病网络的构建用cytoscape软件,并将获得的靶点用DAVID数据库进行GO功能富集及KEGG通路分析。结果从石榴健胃丸中筛选出有效成分96个和366个潜在靶点,其中有44个治疗胃肠炎的关键靶点。KEGG通路分析筛选得到63条信号通路(P <0.05),GO功能富集分析得到GO条目数133个(P <0.05)。在药物-成分-靶点-疾病网络中,显示槲皮素、红花醌甙、杨梅素、β-石竹烯、豆蔻明等成分发挥着重要作用。结论石榴健胃丸治疗胃肠炎疾病的机理可能是通过细胞因子-受体相互作用通路、TNF信号通路和JAK-STAT信号通路等作用于TNF、IL-6、IL-1β和IL-10等靶点,从而发挥抗炎和免疫调节等作用。 Objective To explore the effects targets and signal pathways of Shiliu Jianwei pills, and elaborate its possible mechanisms in the treatment of gastroenteritis. Methods The active ingredients and targets in Shiliu Jianwei pills were screened from the TCMSP and TCMID database, under the conditions of OB ≥ 30% and DL ≥ 0.18.Gastroenteritis targets were obtained from the GeneCards and DisGeNET databases. The protein interaction network was constructed through the String database. The cytoscape software was used to construct the component-target-disease network, and the obtained targets were used in the DAVID database for GO function enrichment and KEGG pathway analysis. Results Shiliu Jianwei Pill contained 96 active ingredients and 366 potential targets. There were 44 potential targets to treat gastroenteritis. KEGG pathway analysis yielded 63 signal pathways(P < 0.05), and GO function enrichment analysis obtained 133 GO entries(P < 0.05). Construction of a drug-component-target-disease network indicated that five compounds(e. g. quercetin, carthamone, myricetin, beta-caryophyllene, cardamomin) played an important role in the entire network. Conclusion The mechanism of Shiliu Jianwei pills for gastroenteritis may be through cytokine-receptor interaction pathway, TNF signaling pathway and JAK-STAT signaling pathway, which acted on TNF, IL-6, IL-1 B and IL-10 targets. So Shiliu Jianwei pills played the role of anti-inflammatory and immune regulation.
