路易斯安娜鸢尾‘Noble Moment’组培快繁体系建立 被引量:3

Establishment of Tissue Culture and Rapid Propagation System for Louisiana Iris'Noble Moment'

摘要 以路易斯安娜鸢尾‘Noble Moment’新生根茎为外植体,以MS为基础培养基,添加不同水平生长素与细胞分裂素为不同的处理,并对组培苗进行移栽试验,探索建立路易斯安娜鸢尾‘Noble Moment’组培快繁体系。结果表明:诱导根茎不定芽的最佳培养基为MS+2.0 mg/L 6-BA+0.2 mg/L NAA和MS+1.0~2.0 mg/L 6-BA+0.5 mg/L KT+0.2 mg/L NAA,诱导率在48.8%~52.6%;利用不定芽诱导丛生芽的最佳培养基为MS+1.0 mg/L 6-BA+0.5 mg/LKT+0.2 mg/LNAA,诱导率达70.4%;利用丛生芽诱导不定根的最佳培养基是1/2MS+0.8 mg/LNAA,诱导率达76.8%。幼苗移栽的最佳基质配比为草炭:珍珠岩:蛭石=3:2:1,幼苗根系发达、生长健壮、叶色浓绿。本研究为路易斯安娜鸢尾的工厂化育苗提供了理论支撑与方法依据,具有现实和深远的意义。 The new rhizome of Louisiana Iris'Noble Moment'is used as explants,with MS as the basic medium,different levels of auxin and cytokinin were added for different treatments,and the tissue culture seedlings were transplanted to explore the establishment of the Louisiana Iris'Noble Moment'tissue culture and rapid propagation system.The results show that the best culture medium for inducing adventitious buds from rhizome is MS+2.0 mg/L 6-BA+0.2 mg/L NAA and MS+1.0~2.0 mg/L 6-BA+0.5 mg/L KT+0.2 mg/L NAA,the induction rate is 48.8%~52.6%.The best culture medium for tufted buds induced by adventitious buds is MS+1.0 mg/L 6-BA+0.5 mg/LKT+0.2 mg/L NAA,the induction rate is 70.4%.The best medium for inducing adventitious roots from clustered buds is 1/2MS+0.8 mg/L NAA,the induction rate is 76.8%.The best substrate ratio for transplanting seedlings is peat:perlite:vermiculite=3:2:1.The seedlings have well-developed root systems,strong growth,and dark green leaves.This study provides theoretical support and method basis for the factory seedling of Louisiana Iris,which has practical and far-reaching significance.
