摘要
目的:检测circ_XPO1在骨肉瘤患者组织和外周血中的表达水平,探讨其临床意义。方法:qRT-PCR检测39例骨肉瘤患者肿瘤组织、瘤旁组织及外周血中circ_XPO1的表达水平和24例健康志愿者外周血中circ_XPO1的表达水平,绘制ROC曲线分析circ_XPO1对骨肉瘤的诊断价值,多因素回归分析研究circ_XPO1与肿瘤病理指标的相关性,通过生存曲线分析和COX回归分析研究circ_XPO1对骨肉瘤患者预后的影响。结果:骨肉瘤患者外周血中circ_XPO1的表达水平比健康志愿者显著升高,而骨肉瘤患者的肿瘤组织和外周血中circ_XPO1的表达量均比瘤旁组织显著升高。绘制ROC曲线,骨肉瘤患者外周血中circ_XPO1表达水平的AUC为0.8579。根据外周血的circ_XPO1表达水平将39例骨肉瘤患者分为circ_XPO1高表达组和circ_XPO1低表达组,多因素回归分析显示外周血circ_XPO1的高表达与组织学Price分级和肺部转移呈正相关;绘制生存曲线分析,外周血circ_XPO1高表达组的5年总生存率显著降低,COX回归分析结果提示外周血circ_XPO1高表达是影响骨肉瘤患者生存率的独立危险因素。结论:骨肉瘤患者的外周血和肿瘤组织中circ_XPO1的表达上调,外周血中circ_XPO1的高表达对骨肉瘤有较高的诊断价值,并且circ_XPO1上调越明显,提示临床病情越严重,预后越差。
Objective:To detect the expression level of circ_XPO1 in tissues and peripheral plasma of patients with osteosarcoma,and explore its clinical significance.Methods:qRT-PCR was used to detect the expression level of circ_XPO1 in tumor tissues,adjacent tissues and peripheral plasma of 39 patients with osteosarcoma,and the expression level of peripheral plasma circ_XPO1 in 24 healthy volunteers.ROC curve was drawn to analyze the diagnostic value of circ_XPO1 for osteosarcoma.Multivariate regression analysis was used to study the correlation between circ_XPO1 and tumor pathological indicators.Survival curve analysis and COX regression analysis was used to study the effect of circ_XPO1 on the prognosis of patients with osteosarcoma.Results:The expression level of circ_XPO1 in peripheral plasma of osteosarcoma patients was significantly higher than that of healthy volunteers.Compared with adjacent tissues,the expression level of circ_XPO1 in tumor tissues and peripheral plasma of osteosarcoma patients was significantly increased.The AUC of the peripheral plasma circ_XPO1 expression of osteosarcoma patients in ROC curve was 0.8579.According to the expression level of peripheral plasma circ_XPO1,39 patients with osteosarcoma were divided into circ_XPO1 high expression group and circ_XPO1 low expression group.Multivariate regression analysis showed that the high expression of peripheral plasma circ_XPO1 was positively correlated with histological Price grade and lung metastasis.Survival curve analysis showed that the 5-year overall survival rate of the high expression group of peripheral plasma circ_XPO1 was significantly reduced.COX regression analysis indicated that the high expression of peripheral plasma circ_XPO1 was an independent risk factor affecting the survival of osteosarcoma patients.Conclusion:The expression of circ_XPO1 in peripheral plasma and tumor tissues of osteosarcoma patients is up-regulated.The high expression of peripheral plasma circ_XPO1 has a good diagnostic value for osteosarcoma.The more up-regulated circ_XPO1 expression,the worse the clinical condition,and the worse the prognosis.
