摘要
目的探讨间充质上皮转化因子(mesenchymal epithelial transition factor,MET)作为胰腺癌患者临床预后评价指标的意义。方法从GEO数据库下载GSE28735和GSE62452基因芯片,应用生物信息学分析MET基因在胰腺癌组织及其癌旁组织中的表达差异;从TCGA数据库下载胰腺癌基因芯片分析MET基因表达与胰腺癌患者临床病理特征的相关性及预后风险;最后,通过GO和KEGG富集分析MET参与胰腺癌发生的可能分子机制。结果MET基因在胰腺癌组织中的表达水平高于癌旁组织(P<0.001);MET基因高表达组胰腺癌患者的总体生存率和无病生存率均低于低表达组(均P<0.001);MET基因表达水平与胰腺癌患者的年龄、肿瘤T分期以及肿瘤的组织学分级有关(P<0.05),并且MET基因高表达、患者年龄>65岁和肿瘤N1分期是影响胰腺癌患者预后的独立危险因素。KEGG富集分析结果显示,MET主要与PI3K/AKT信号通路、FAK信号通路、癌症转录失调节等途径有关。结论MET可能是胰腺癌有价值的肿瘤标志物,并可预测胰腺癌患者的不良预后。
Objective To explore the significance of mesenchymal epithelial transition factor(MET)as a clinical prognostic evaluation index for patients with pancreatic cancer based on bioinformatics analysis.Methods The GSE28735 and GSE62452 gene chips from GEO database were downloaded and the difference of MET gene expression between cancer and adjacent cancerous tissues were analyzed by bioinformatics.We downloaded pancreatic cancer gene chip from TCGA database to analyze the correlation between MET gene expression and clinicopathological features of pancreatic cancer patients and prognosis risk.Finally,the possible molecular mechanism of MET involved in pancreatic carcinogenesis was analyzed by GO and KEGG enrichment analysis.Results The expression level of MET gene in pancreatic cancer tissues was significantly higher than that in adjacent cancerous tissues(P<0.001).The overall survival and disease-free survival of pancreatic cancer patients in the high MET gene expression group were lower than those in the low expression group(P<0.001).The expression level of MET gene was related to the age of pancreatic cancer patients,T stage,and histological grading of tumors(P<0.05),and high MET gene expression,age>65 years,and N1 stage were independent risk factors affecting the prognosis of pancreatic cancer patients.KEGG enrichment analysis showed that MET was mainly related to PI3 K/AKT signaling pathway,FAK signaling pathway,and cancer transcription dysregulation and so on.Conclusion MET may be a valuable tumor marker for pancreatic cancer and can predict the poor prognosis of patients with pancreatic cancer.
