油桐花水提物对烧伤大鼠组织蛋白酶B、D的影响

摘要 目的:油桐花水提物对烧伤有较好的治疗作用,在此基础上进一步探索对烧伤大鼠组织蛋白酶B、D的影响情况。方法:选60只大鼠均模拟烧伤,烧伤部分做组织切片HE染色观察烧伤造模情况,分成空白组、拟烧伤组、阳性组和水提物高、中、低浓度组,分别对六组大鼠涂抹三个浓度水提物,通过免疫组化法、Elisa和Western Blot法对Cathepsin B、Cathepsin D检测含量以及表达情况。结果:烧伤大鼠的Cathepsin B以及Cathepsin D含量明显多于空白组(P<0.01)。免疫组化法结果显示,中高浓度水提物对烧伤大鼠组织蛋白酶B有显著抑制作用(P<0.01),对组酶D表达有抑制作用(P<0.05)。Western Blot法结果中,与烧伤组相比,高浓度水提物会明显下调Cathepsin B表达(P<0.01)。ELisa法结果显示中高浓度水提物对组织蛋白酶D明显抑制(P<0.01)。结论:烧伤大鼠组织蛋白酶B、D表达明显升高,中高浓度的油桐花水提物对于烧伤大鼠的组织蛋白酶B、D有明显的抑制作用。 Objective:The water extract of Vernicia fordii(Hemsl.)has a good therapeutic effect on burns.For further exploration,the water extract of Vernicia fordii(Hemsl.)on cathepsin B and D of burn rats is carried out.Methods:Sixty rats were selected to simulate burns,and the burned parts were treated with HE staining to observe the burn model.They were divided into blank group,simulated burn group,positive group and high,medium and low concentration groups of water extract,respectively.The rats were smeared with three concentrations of water extracts,and the content and expression of Cathepsin B and Cathepsin D were detected by immunohistochemistry,Elisa and Western Blot methods.Results:The contents of Cathepsin B and Cathepsin D in burn rats were significantly higher than those in the blank group(P<0.01).The results of immunohistochemistry showed that the medium and high concentration water extracts had a significant inhibitory effect on cathepsin B in burn rats(P<0.01),and inhibited the expression of group enzyme D(P<0.05).For Western Blot method,the high-concentration water extract significantly down-regulated the expression of Cathepsin B(P<0.01)compared with the burn group.For ELisa method,the medium and high concentration of water extracts significantly inhibited cathepsin D(P<0.01).Conclusions:The expression of cathepsin B and D of burned rats was significantly increased,and medium and high concentrations of water extract of V.fordii had a significant inhibitory effect on cathepsin B and D of burned rats.
