摘要
目的观察RNA干扰(RNAi)介导的CD151基因沉默对子宫颈癌细胞生长、迁移及侵袭的影响。方法采用免疫组化EnVision法检测子宫颈鳞状细胞癌及慢性子宫颈炎组织中CD151蛋白的表达;设计并筛选抑制CD151 mRNA表达的siRNA序列,构建含有shRNA序列的质粒转染子宫颈癌细胞;采用Western blot法筛选稳定沉默CD151基因的子宫颈癌细胞株,用于后续的细胞增殖、周期、凋亡、迁移和侵袭实验。结果CD151蛋白在子宫颈鳞状细胞癌组织中阳性;与空载体组及空白对照组相比,转染Psciencer2.0-shRNA-CD151重组质粒的子宫颈癌细胞CD151表达水平显著下降,细胞增殖活力明显降低,细胞周期被阻滞在G 1期,细胞凋亡显著增加,细胞迁移和侵袭能力亦受到明显抑制,差异有统计学意义(P均<0.05)。结论通过RNAi介导的CD151基因沉默可明显抑制子宫颈癌细胞的生长及恶性生物学行为,CD151有望成为治疗子宫颈癌的潜在靶点。
Purpose To study the effect of RNA interference(RNAi)-mediated CD151 gene silencing on the growth,migration and invasion of cervical cancer cells.Methods Expression of CD151 in cervical squamous cell carcinoma and chronic cervicitis tissues was evaluated by immunohistochemical staining.siRNA sequences were screened and designed that inhibit the expression of CD151 mRNA,the plasmid containing the sequence shRNA was constructed to transfect cervical cancer cells.Western blot was used to screen cervical cancer cell lines that have constantly silenced CD151 gene.The selected cervical cancer cells were subjected to subsequent cell proliferation,cycle,apoptosis,migration and invasion experiments.Results CD151 was positively expressed in the cell membrane or cytoplasm of cervical squamous cell carcinoma tissues.Compared with the blank vector group and the blank control group,the expression of CD151 in cervical cancer cells infected with Psciencer2.0-shRNA-CD151 significantly decreased,cell proliferation activity was significantly reduced,cell cycle was blocked in the G_(1)phase,apoptosis was significantly increased,cell migration and the invasion ability were also significantly suppressed(P<0.05).Conclusion RNAi-mediated CD151 gene silencing can significantly inhibit cervical cancer cell growth and malignant biological behavior.CD151 exhibits a potential target for the treatment of cervical cancer.
