摘要
为探究鸡源禽腺病毒的适应细胞及培养特性,将前期分离保存的1株4型禽腺病毒滨州株,分别接种Vero细胞、QT-35细胞、DF-1细胞和LMH细胞,连续传3代后,通过细胞病变特征和病毒含量,选出最适宜的细胞,并对细胞进行克隆优选。同时研究了该毒株的接毒剂量、接毒后吸附时间和收获时间。结果表明,LMH细胞最适于4型禽腺病毒滨州株的培养。最佳增殖条件为:最佳接毒量为1/1000、接毒后最佳吸附时间为1 h、收毒时间为接毒后36 h。
To explore the adaptive cells and the culture characteristics of the fowl adenovirus,a serotype 4 fowl adenovirus Binzhou strain isolated previously were inoculated into the Vero cell,QT-35 cell,DF-1 cell and LMH cell.After three successive generations,the most suitable cells were selected according to the characteristics of cytopathic changes and virus content,and the suitable cells were selected for cloning and optimization.At the same time,the amount of inoculated virus,adsorption time and harvest time of this virus strain were studied.The results showed that LMH cells are most suitable for the culture of type 4 fowl adenovirus Binzhou strain.The optimal proliferation condition to cultivate the virus train is obtained as follows:the inoculum concentration is 1/1000(v/v),the adsorption time is 1 h after inoculation,and the harvest time is 36 h after inoculation.
