摘要
通过诱变得到枯草芽孢杆菌SYL-1,具有较高的核黄素产量。发酵测定其核黄素产量,在42 h达到1.90 g/L(±0.020 g/L)。利用实时荧光定量PCR系统分析其核黄素素代谢通路上基因表达量的变化,其磷酸戊糖途径和嘌呤代谢途径上的基因gmk、gntZ、prs、yqeC、yqjI、zwf以及嘌呤操纵子上的基因purE表达量相较于构建的菌株LY-05有所提高。在LY-05中,过表达基因prs、purF、gntZ、zwf核黄素产量提高。通过解析SYL-1菌株核黄素代谢通路上基因表达量的变化,找出影响枯草芽孢杆菌核黄素产量的关键位点。
Bacillus subtilis SYL-1 with high riboflavin yield was obtained by mutation.The riboflavin yield was determined by fermentation and reached 1.90 g/L(±0.020 g/L)in 42 h.The changes of gene expression in riboflavin metabolic pathway were analyzed by real-time fluorescence quantitative PCR system.The expression of gene gmk,gntz,prs,yqeC,yqjI,zwf in pentose phosphate pathway and purine metabolic pathway and purE gene in purine operon were higher than those in strain LY-05.The yields of overexpression genes prs,purF,gntZ,zwf riboflavin increased in LY-05.The key sites affecting riboflavin production of Bacillus subtilis were found by analyzing the changes of gene expression in riboflavin metabolic pathway of SYL-1 strain.
