桦褐孔菌发酵条件的优化及体外降糖作用的研究 被引量:2

摘要 [目的]研究桦褐孔菌的潜在应用价值,并分析其菌丝体多糖的降糖作用。[方法]以生物量和粗多糖含量为指标,通过单因素试验与正交试验设计方法对其发酵条件进行优化。将桦褐孔菌菌丝多糖对糖尿病模型小鼠进行灌胃,测定小鼠血糖变化。[结果]促进该菌株发酵生长的最佳碳源、氮源分别为葡萄糖和牛肉膏;最佳培养基组成为葡萄糖30 g/L,蛋白胨4 g/L,牛肉膏8 g/L。摇床培养条件为pH 7.0,装液量90 mL/250 mL,接种量15%,发酵时间12 d。与模型组相比,桦褐孔菌菌丝多糖能显著降低糖尿病小鼠血糖。[结论]该研究可为工业化生产桦褐孔菌多糖及实际应用提供参考。 [Objective]To evaluate the potential application value of Inonotus obliquus and analyze the hypoglycemic effect of mycelium polysaccharide.[Method]Using biomass and crude polysaccharide yield as indicators,the fermentation conditions of Inonotus obliquus were optimized by single factor and orthogonal experiments.The diabetic model mice were intragastrically inoculated with Inonotus obliquus mycelium polysaccharide,and blood glucose changes in the mice were measured.[Result]The results showed that the most suitable carbon source and nitrogen source are glucose and beef extract,respectively.The optimum fermentation conditions were 30 g/L glucose,4 g/L peptone,8 g/L beef extract.Table concentrator culture conditions were pH 7.0,loaded liquid 90 mL/250 mL,inoculation amount 15%and fermentation time 12 d.Compared with the model group,Inonotus obliquus mycelium polysaccharide can significantly reduce blood glucose in diabetic mice.[Conclusion]The study can provide reference for the industrial production of Inonotus obliquus polysaccharide and its practical application.
